摘要
目的探讨厚朴酚对鼻咽癌细胞增殖和侵袭的影响及其潜在的机制。方法采用CCK-8检测细胞的活性;集落形成试验检测细胞的生长;caspase-3和caspase-9活性试剂盒分别检测细胞的caspase-3和caspase-9的活性;Transwell侵袭试验检测细胞的侵袭能力;实时定量PCR检测细胞中N-cadherin和vimentin mRNA的表达水平。结果厚朴酚浓度为50、100以及200μM时能够显著抑制HK1和CNE2细胞的活性,与对照组(0μM)比较,差异具有统计学意义(P<0.05);厚朴酚对HK1和CNE2细胞的抑制呈浓度依赖性。厚朴酚(100μM)显著抑制HK1和CNE2细胞集落形成数量,同时提高细胞中caspase-3和caspase-9的活性,与对照组比较,差异具有统计学意义(P<0.05)。厚朴酚(100μM)能够显著降低鼻咽癌细胞的侵袭数量,与对照组比较,差异显著(P<0.05)。同时,厚朴酚(100μM)能够下调鼻咽癌细胞中N-cadherin和vimentin的表达,与对照组比较,差异具有统计学意义(P<0.05)。结论厚朴酚能够抑制鼻咽癌细胞的增殖和侵袭,提高caspase-3和caspase-9活性以及下调N-cadherin和vimentin的表达。
Objective To investigate the effects of magnolol on the proliferation and invasion of nasopharyngeal carcinoma cells and its underlying mechanism.Methods CCK-8 assay was used to detect cell viability;colony forming assay was used to detect cell growth;caspase-3 and caspase-9 activity kits were used to detect caspase-3 and caspase-9 activities;Transwell invasion test was used to detect the invasion ability of cells;real-time quantitative PCR was used to detect the expression levels of N-cadherin and vimentin mRNA.Results Magnolol significantly inhibited the activity of HK1 and CNE2 cells at concentrations of 50,100 and 200μM,and the differences were statistically significant compared with the control group(0μM)(P<0.05);the inhibition of magnolol on HK1 and CNE2 cells was concentration dependent.Magnolol(100μM)significantly inhibited the colony formation of HK1 and CNE2 cells,and increased the activities of caspase-3 and caspase-9 in the cells,and the differences were statistically significant compared with the control group(P<0.05).Magnolol(100μM)can significantly reduce the number of nasopharyngeal carcinoma cell invasion,and the difference was statistically significant compared with the control group(P<0.05).At the same time,magnolol(100μM)could down-regulate the expression of N-cadherin and Vimentin in nasopharyngeal carcinoma cells,and the difference was statistically significant compared with the control group(P<0.05).Conclusion Magnolol can inhibit the proliferation and invasion of nasopharyngeal carcinoma cells,increase the activity of caspase-3 and caspase-9,and down-regulate the expression of N-cadherin and vimentin.
