摘要 目的通过网络药理学方法探讨黄连治疗动脉粥样硬化(AS)的作用机制。方法利用TCMSP数据库获取黄连的主要化学成分。借助DisGeNET、GeneCards数据库获取AS相关的潜在作用靶点,通过Cytoscape V3.7.2软件构建“药物-活性成分-靶蛋白-疾病”网络;采用String数据库建立蛋白互作网络(PPI),使用DAVID数据库分析基因本体(GO)功能及基因组百科全书(KEGG)通路。结果筛选黄连有效成分14个化学成分和128个靶点,与AS相关的交集靶点62个;“药物-活性成分-靶蛋白-疾病”网络中Degree排前3的成分为槲皮素、巴马汀、小檗碱;GO功能注释与KEGG通路富集分析共得到108个GO注释,40条KEGG通路,按P值大小,筛选出靠前20个GO注释和KEGG富集通路。结论黄连治疗AS涉及多个靶点、生物学过程和信号通路,为其药理机制研究提供新的思路和理论参考。 Objective To explore the mechanism of in treating Atherosclerosis by network pharmacology.Method The TCMSP database was used to obtain the main chemical components of Coptis Chinensis.The potential targets related to Atherosclerosis were obtained by DisGeNET and GeneCards databases and the network of drug-active ingredient-target protein-diseases was built by Cytoscape V3.7.2 software.String database was used to build a protein interaction network(PPI)and DAVID database was used to analyze GO function and KEGG pathway.Results 14 chemical components and 128 targets were extracted from Coptis Chinensis,in which 62 intersecting targets related to Atherosclerosis.The top 3 components of Degree in the drug-active component-target protein-diseases network are Quercetin,Palmatine,and Berberine.108 GO annotations and 40 KEGG pathways were obtained by analyzing GO functional annotation and KEGG pathway enrichment.The top 20 GO annotations and KEGG enrichment pathways were selected according to the P value.Conclusions Coptis Chinensis’treatment of Atherosclerosis involves multiple targets,biological processes and signal pathways,which provide new ideas and theoretical references for the study of its pharmacological mechanisms.
