生大黄粉治疗新生鼠坏死性小肠结肠炎实验研究

Experimental Study on the Treatment of Necrotizing Enterocolitis in Neonatal Rats with Rhubarb Powder

目的观察生大黄粉对坏死性小肠结肠炎模型新生鼠肠组织中胃动素(MTL)和促胃液素(GAS)含量的影响,探讨其治疗机制,为临床应用生大黄粉治疗新生儿NEC提供科学依据。方法将72只新生2d龄SD大鼠随机分为空白对照组,生理盐水组,西咪替丁组,生大黄粉大、中、小剂量组,每组12只,除空白对照组外其余各组建立NEC模型,同时按要求予药物治疗,第4天空腹颈椎脱臼处死,取回盲部近端肠组织做病理切片,剩余肠组织制成组织匀浆,用于检测肠组织MTL及GAS含量。结果与生理盐水组比较,生大黄粉治疗组MTL升高、GAS降低,差异均有统计学意义(P<0.05);与西咪替丁组比较,生大黄粉治疗组中MTL表达高于西咪替丁组,GAS表达低于西咪替丁组,差异均有统计学意义(P<0.05);生大黄粉各剂量组之间比较,MTL、GAS表达相似,差异均无统计学意义(P>0.05)。结论生大黄粉能够降低新生大鼠NEC模型肠损伤程度,且效果优于西咪替丁。

Objective To explore the effects of raw rhubarb powder on the contents of motilin (MTL) and gastric (GAS) in the intestinal tissues of NEC rats,and to explore the therapeutic mechanism,so as to provide scientific basis for clinical application of rhubarb in the treatment of newborn NEC.Methods 72 newborn SD rats were randomly divided into 2d age blank control group,saline group and cimetidine group,raw rhubarb powder large,medium and small dose group,each group of 12,establishes a model of NEC groups except the blank control group,at the same time,according to the requirements given drug treatment,4d cervical dislocation executed on an empty stomach,retrieve the blindness of the proximal intestinal tissue pathological slices,residual intestinal tissue made from tissue homogenate,used to detect intestinal tissue MTL and GAS content.Results Compared with normal saline group,MTL increased and GAS decreased in rhubarb powder treatment group,with statistically significant differences (P<0.05).Compared with the cimetidine group,MTL expression in the raw rhubarb powder treatment group was higher than that in the cimetidine group,and GAS expression was lower than that in the cimetidine group,with statistically significant differences (P<0.05).MTL and GAS expressions were similar between groups of raw rhubarb powder with no statistical significance (P>0.05).Conclusion rhubarb powder can reduce the degree of intestinal injury in NEC rats,and it is better than cimetidine.
