基于L-天冬氨酸功能化纳米金比率型探针的卡那霉素A比色法检测 被引量:4

A ratiometric L-aspartic acid functionalized gold nanoparticles probe for colorimetric detection of kanamycin A

摘要 设计基于L-天冬氨酸(L-asp)功能化的金纳米粒(Asp-AuNPs)比率型探针,用于尿液中卡那霉素A(Kana A)的比色法测定。以氯金酸(HAuCl4·3H2O)为金源前驱体、硼氢化钠(NaBH4)为还原剂、L-asp为稳定剂一步法制备Asp-AuNPs复合物,优化溶液p H、L-asp和NaBH4浓度等制备条件,获得灵敏高、稳定性好、抗干扰能力强的Asp-AuNPs探针,探讨了比色响应的机理。结果显示,在50~500 nmol/L范围内,吸光度比值(A690/A525)与Kana A浓度之间呈良好的线性相关(r=0. 998),最低检出限为28. 3 nmol/L(16. 5μg/L)。尿液中共存物质对Kana A检测不产生干扰,回收率范围为92. 8%~108. 2%。该方法可准确检测尿液中的Kana A,为临床尿液中Kana A的浓度监测提供了一种新的分析手段。 A L-aspartic acid( L-asp) functionalized gold nanoparticles( Asp-AuNPs) ratiometric probe was synthesized for colorimetric detection of kanamycin A( Kana A) in urine samples. One-step preparation of AspAu NPs composite was realized by using chloroauric acid as precursor,sodium borohydride( NaBH4) as reducing agent and L-asp as capping agent. Optimization of the preparation conditions including the media pH,concentration of L-asp and NaBH4 led to the formation of sensitive,selective and stable Asp-AuNPs probe. In addition,the possible colorimetric response mechanism was also proposed. A good linear correlation was established between the A690/A525 values with the analyte concentration in the range of 50 ~ 500 nmol/L( r =0. 998),and the limit of detection( LOD) for Kana A was estimated to be 28. 3 nmol/L( 16. 5 μg/L). The coexisting substances in urine showed negligible interfering effect to the analysis of Kana A with the recoveries ranging from 92. 8% to 108. 2%. This method offers a possibility for monitoring Kana A concentration level in clinical samples.
