基于网络药理学研究益智仁治疗阿尔兹海默症的物质基础与作用机制 被引量:4

Network Pharmacology-based Study on Material Basis and Mechanism of Alpinia Oxyphyllain Treatment of Alzheimer's Disease

摘要 目的:采用网络药理学方法分析中药益智仁治疗阿尔兹海默症(AD)的药效物质基础及其分子作用机制。方法:通过TCMSP及TCMID数据库及查阅文献筛选出益智仁的潜在活性成分,运用TCMSP与Swiss Target Prediction数据库预测其作用靶点;检索Gene Cards和OMIM数据库获得AD的相关靶基因;绘制韦恩图获得"益智仁-AD"共有靶点。使用Cytoscape软件构建"益智仁-潜在活性成分-相关靶点-AD"相互作用网络;利用Bioconductor工具对共有靶点进行GO功能注释和KEGG通路富集分析。结果:共获得益智仁抗AD潜在有效成分10个,治疗AD相关基因8435个,这些成分共作用于158个靶点,其中有113个靶点与AD相关。GO分析结果显示,益智仁治疗AD的生物过程主要涉及蛋白丝氨酸激酶活性、裂解酶活性、碳氧裂解酶活性等;KEGG通路富集分析结果显示,益智仁治疗AD调控的信号通路主要涉及PI3K-Akt信号通路、Ras信号通路等。结论:益智仁中白杨素、原儿茶酸、圆柚酮和豆甾醇等活性成分可能是其治疗AD的主要物质基础之一,主要通过抗炎、抗氧化、抗凋亡、促进细胞增殖分化和提高免疫力等方面治疗AD。研究结果为益智仁抗AD物质基础及作用机制的阐明提供了一定的理论依据,并为中药防治AD的机制研究提供参考思路。 Objective To analyze the material basis and mechanism of Alpinia oxyphyllain the prevention and treatment of Alzheimer s Disease(AD).Methods TMTSP database,TCMID database and literature mining were used to screen Alpinia oxyphylla active ingredients,TCMSP database and SwissTargetPrediction database were used topredictits targets.Gene Cards and OMIM databases were retrieved to obtain the relevant target genes of AD.Draw the Venn diagram to obtain the common target of Alpinia oxyphylla-AD.The interaction network of Alpinia oxyphylla-potential active ingredients-related targets–AD was constructed by Cytoscape software.GO functional annotation and KEGG pathway enrichment analysis were performed on common targets using Bioconductor tools.Results A total of 8435 AD related genes and 10 potential effective components of Alpinia oxyphylla against AD were obtained,which acted on 158 targets,113 of which were related to AD.Go analysis results showed that the biological process of potential genes of Alpinia oxyphylla treating AD mainly involved protein serine kinase activity,lyase activity,carbonyl-oxygen lyase activity,etc.KEGG pathway enrichment analysis showed that the potential regulatory signal pathways of Alpinia oxyphylla in treating AD mainly involved Alzheimer disease,PI3K-Akt signaling pathways,Ras signaling pathways,etc.Conclusion The active components of Alpinia oxyphylla,such as chrysin,protocatechuic acid,nootkatone and stigmasterol,may be thematerial basis for the treatment of AD,mainly throughanti-inflammatory,anti-oxidation,anti-apoptosis,promoting cell proliferation and differentiation and improving immunity.Network pharmacology provides scientific basis for elucidating the anti-AD mechanism of Alpinia oxyphylla,and provides a reference for the research on the prevention and treatment mechanism of traditional Chinese medicine.
