摘要
目的:建立一种太子参环肽单体的分离制备方法。方法:采用硅胶柱层析分离制备太子参环肽粗分物,经制备级高效液相色谱(HPLC)纯化,使用质谱(MS)、核磁共振波谱(NMR)及二维核磁共振波谱对其结构进行表征。结果:通过质谱、核磁共振波谱法确定了所得环肽为太子参环肽Pseudostellaria A (PA)。结论:建立一种太子参环肽PA的分离制备方法,提取率为32. 5μg/g,纯度为95. 16%。
Objective To establish a method for the separation and preparation of Pseudostellaria heterophylla cyclic peptide monomer.Methods The crude cyclic peptide of Pseudostellaria heterophylla was separated and prepared by silica gel column chromatography,purified by preparative High Performance Liquid Chromatography(HPLC),and its structure was characterized by Mass spectrometry(MS),Nuclear Magnetic Resonance spectroscopy(NMR)and two-dimensional nuclear magnetic resonance spectroscopy.Results The obtained cyclic peptide was determined to be Pseudostellaria A(PA)by mass spectrometry and nuclear magnetic resonance spectroscopy.Conclusion A method for the separation and preparation of Pseudostellaria heterophylla cyclic peptide PA has been established,the extraction rate is 32.5μg/g,and the purity is 95.16%.
