热敏灸对慢性萎缩性胃炎大鼠Ghrelin及GHSR的影响 被引量:8

Effects of heat-sensitive moxibustion on Ghrelin and GHSR in rats with chronic atrophic gastritis

摘要 目的:观察热敏灸对慢性萎缩性胃炎(CAG)大鼠生长激素释放肽(Ghrelin)及生长激素促分泌素受体(GHSR)的影响,探讨其可能的作用机制。方法:66只雄性SD大鼠,采用随机数字表法随机分为空白组、造模组。空白组不予干预,造模组采用复合病因造模法造模12周,随机分为模型组、维酶素组、艾灸组,艾灸组艾灸中脘穴40min,艾灸后按2mL·kg^-1·d^-1灌服0.9%氯化钠溶液,根据艾灸过程中大鼠尾温变化分为热敏灸组和非热敏灸组,维酶素组灌服等量维酶素,1次/d,共28d。观察各组大鼠一般状况、体质量等变化,Western blot法检测下丘脑、海马组织中Ghrelin、GHSR蛋白表达,光镜观察胃黏膜组织病理学变化。结果:①造模后,造模组大鼠表现为精神不振,活动迟钝,饮食量下降,毛发枯槁,背部松垮,大便质软或稀溏等;体质量增长缓慢。②治疗后,与空白组比较,模型组体质量仍差距较大(P<0.01);与模型组比较,非热敏灸组、热敏灸组和维酶素组体质量均明显增长(P<0.05)。③与空白组比较,模型组大鼠下丘脑及海马组织中Ghrelin及GHSR蛋白表达均减少(P<0.01);与模型组比较,热敏灸组、维酶素组、非热敏灸组均可上调下丘脑及海马组织中的Ghrelin及GHSR蛋白表达(P<0.01),热敏灸组在下丘脑组织中的Ghrelin、GHSR,以及海马组织中的GHSR蛋白表达明显优于其他各组(P<0.05,P<0.01)。结论:热敏灸可改善CAG大鼠的胃肠动力及胃黏膜损伤,从而达到治疗CAG的目的,这可能与其增加下丘脑及海马组织中的Ghrelin及GHSR蛋白表达水平有关。 Objective:To observe the effects of heat-sensitive moxibustion on Ghrelin and its receptor GHSR in rats with chronic atrophic gastritis(CAG),and to explore its possible mechanism for treating CAG.Methods:According to random number table,66 SD male rats were randomly divided into blank group and model group.The rats in the blank group did not intervene,the rats in the model group were treated with the composite etiology model method to establish CAG model for 12 weeks,and they were randomly divided into model group,vitacoenzyme group and moxibustion group.The rats in the moxibustion group were treated with moxibustion at Zhongwan(CV 12)acupoint for 40 min.After moxibustion,the rats were treated with intragastric administration of 0.9%NaCl according to 2 mL·kg^-1·d^-1.According to the change of tail temperature during moxibustion,the rats were divided into heat-sensitive moxibustion group and non heat-sensitive moxibustion group.The rats in the vitacoenzyme group were treated with identical volume of vitacoenzyme,once a day for 28 days.The general condition and body weight of rats in each group were observed.The expressions of Ghrelin and GHSR protein in hypothalamus and hippocampus were detected by Western blot method.Optical microscope was applied to observe the histopathological changes of gastric mucosa.Results:①After model establishment,the rats in the model group were dispirited,activity slower,diet decreased,withered hair,weaked back,soft or watery stool,and slow growth of body weight.②After treatment,compared with the blank group,the body weight in the model group were still significantly different(P<0.01).Compared with the model group,the body weight in the non heat-sensitive moxibustion group,heat-sensitive moxibustion group and vitacoenzyme group were obviously increased(P<0.05).③Compared with the blank group,Ghrelin and GHSR protein expressions in hypothalamus and hippocampus of the model group were decreased(P<0.01).Compared with the model group,the heat-sensitive moxibustion group,vitacoenzyme group and non heatsensitive moxibustion group were up-regulated the expressions of Ghrelin and GHSR protein in hypothalamus and hippocampus(P<0.01).The heat-sensitive moxibustion group in increasing the expressions of Ghrelin and GHSR protein in hypothalamus and the expressions of GHSR protein in hippocampus were significantly superior to the other groups(P<0.05,P<0.01).Conclusion:Heat-sensitive moxibustion can improve gastrointestinal motility and gastric mucosal injury of CAG rats,thus achieving the purpose of treating CAG,which may be related to the increase of Ghrelin and GHSR protein expression levels in hypothalamus and hippocampus.
