新疆5种阿魏对人胃癌SGC-7901细胞体外增殖抑制及凋亡作用研究

Effects of five kinds of Resina Ferulae in Xinjiang on cell proliferation inhibition and apoptosis of human gastric carcinoma SGC-7901 cells in vitro

目的:考察新疆5种阿魏(托里阿魏、新疆阿魏、大果阿魏、阜康阿魏、圆锥茎阿魏)不同的极性部位(乙醇提取物及石油醚、二氯甲烷、乙酸乙酯、水饱和正丁醇、水部位)提取物对人胃癌SGC-7901细胞体外增殖抑制作用和5种阿魏乙醇提取物对人胃癌SGC-7901细胞的凋亡作用。方法:以新疆5种阿魏各极性部位不同浓度(9.375、18.750、37.500、75.000、150.000、300.000mg/L)干预人胃癌SGC-7901细胞,通过四唑盐比色法检测吸光度(OD值),求得细胞增殖抑制率,应用流式细胞术分别检测5种阿魏乙醇提取物的细胞凋亡率。结果:新疆5种阿魏各极性部位均对人胃癌SGC-7901细胞产生增殖抑制作用;托里阿魏、新疆阿魏、大果阿魏的乙醇提取物抑制作用较好,IC50分别为26.06、42.27、28.31mg/L;其中抑制作用较强的部位多集中于石油醚、二氯甲烷、乙酸乙酯部位,水饱和正丁醇和水提部位抑制作用较差。大果阿魏乙醇提取物部位对人胃癌SGC-7901细胞有较明显的的促凋亡作用。结论:结合细胞增殖抑制和凋亡实验结果,大果阿魏作用较明显,考虑其具有一定的抗肿瘤效果,值得更进一步的深入研究。

Objective:To investigate the effects of different polar parts extracts(ethanol extract,petroleum ether,dichloromethane,ethyl acetate,water saturated n-butanol,water part)of five kinds of Resina Ferulaein in Xinjiang(Torri Resina Ferulaein,Xinjiang Resina Ferulaein,Daguo Resina Ferulaein,Fukang Resina Ferulaein and Yuanzhui Resina Ferulaein)on cell proliferation inhibition and apoptosis of human gastric carcinoma SGC-7901 cells in vitro and effects of alcohol extracts from 5 kinds of Resina Ferulae on apoptosis of human gastric carcinoma SGC-7901 cells in vitro.Methods:Different concentrations(9.375,18.750,37.500,75.000,150.000,300.000 mg/L)of the six polar parts of five Resina Ferulae in Xinjiang were used to intervene in the human gastric carcinoma SGC-7901 cells.The absorption degree(OD value)was measured by the tetrazole salt colorimetric method to obtain the cell proliferation inhibition rate.The cell apoptosis rate of five kinds of alcohol extracts was measured by flow cytometry.Results:All the polar parts of the five kinds of Resina Ferulae in Xinjiang produced proliferative inhibition of human gastric carcinoma SGC-7901 cells.The ethanol extracts of Torri Resina Ferulae,Xinjiang Resina Ferulae,and Daguo Resina Ferulae have a good inhibitory effect,the IC50 were 26.06,42.27,28.31 mg/L respectively;Among them,the parts with strong inhibition were mostly concentrated in petroleum ether,dichloromethane,and ethyl acetate.The inhibition of watersaturated n-butanol and water-lifting sites was poor.The site of Daguo Resina Ferulae ethanol extract had a more significant effect on human gastric carcinoma SGC-7901 cells apoptosis.Conclusion:Combined with the results of cell proliferation inhibition and apoptosis,the effect of Daguo Resina Ferulae is more significant,which is considered to have certain anti-tumor effects,it is worth further in-depth study.
