基于线粒体途径研究知柏地黄汤对解脲脲原体感染大鼠生精细胞活力的影响 被引量:4

Effects of Zhibai Dihuang Decoction on spermatogenic cell activity in ureaplasma urealyticum infected rats based on mitochondrial pathway

摘要 目的:基于线粒体途径探讨知柏地黄汤含药血清干预解脲脲原体(UU)感染大鼠生精细胞活力的影响及其作用机制。方法:采用在体UU感染建模,随后原代提取大鼠生精细胞,以知柏地黄汤含药血清(低、中、高剂量组)为实验施加因素,同时设空白组、强力霉素组和模型组。给予相应药物干预后进行指标检测。结果:知柏地黄汤含药血清能改善UU感染的生精细胞活性,提高存活率(P<0.01,P<0.05);知柏地黄汤含药血清能有效提高UU感染的生精细胞线粒体ANT4转运活性(P<0.01,P<0.05);知柏地黄汤含药血清能有效上调UU感染的生精细胞中VDAC2、Cyp-D蛋白和mRNA表达水平(P<0.01,P<0.05),下调VDAC1蛋白和mRNA表达水平(P<0.01,P<0.05)。结论:知柏地黄汤可能通过抑制UU感染引起的VDAC1的过度激活,上调VDAC2、Cyp-D mRNA和蛋白表达,上调ANT4的转运活性,进而明显改善线粒体结构的破坏,最终改善生精细胞的活力。 Objective:Based on the mitochondrial pathway,the effects of Zhibai Dihuang Decoction containing serum on apoptotic cells of spermatogenic cells in ureaplasma urealyticum(UU)infected rats and its mechanism were studied.Methods:UU infection in vivo was modeled,and spermatogenic cells of rats were extracted in the primary generation.Serum containing Zhibai Dihuang Decoction(low,medium and high dosage groups)was used as the experimental application factor,while blank group,doxycycline group and model group were set up.After giving the corresponding drug intervention,the test was carried out.Results:The serum containing Zhibai Dihuang Decoction improved the activity of spermatogenic cells infected by UU and increase the survival rate(P<0.01,P<0.05).The serum containing Zhibai Dihuang Decoction effectively improved the mitochondrial ANT4 transport activity of spermatogenic cells infected with UU(P<0.01,P<0.05).The serum containing Zhibai Dihuang Decoction effectively up-regulated the expression of VDAC2 and Cyp-D protein and mRNA in spermatogenic cells infected with UU(P<0.01,P<0.05),and down-regulated the expression of VDAC1 protein and mRNA(P<0.01,P<0.05).Conclusion:Zhibai Dihuang Decoction can significantly improve the damage of mitochondrial structure,repair and protect mitochondria by inhibiting the over-activation of VDAC1 caused by UU infection,up-regulating the expression of VDAC2 and Cyp-D,and up-regulating the transporting activity of ANT4,and ultimately improving the viability of spermatogenic cells.
