刺山柑总生物碱对系统性硬皮病小鼠Wnt10b、Dickkopf-1及E-钙粘蛋白的影响 被引量:3

Effects of capparis spinosa total alkaloid on Wnt10b,Dickkopf-1 and E-Cadherin expressions in mice with systemic sclerosis

摘要 目的:观察刺山柑总生物碱对系统性硬皮病(SSc)Wnt通路相关蛋白Wnt10b、Dickkopf-1(DKK-1)及E-钙粘蛋白(E-Cadherin)的影响。方法:BALB/c小鼠SSc模型的复制采用博莱霉素皮下注射法,然后给予受试药乳膏(225、450、900mg/kg)治疗8周。取小鼠注射区皮肤,通过Western blot检测α-平滑肌肌动蛋白(α-SMA)表达,ELISA实验检测Wnt10b和E-Cadherin含量;取血清,ELISA实验检测DKK-1含量。结果:刺山柑总生物碱225、450、900mg/kg可明显降低SSc小鼠α-SMA和Wnt10b表达(P<0.01),升高DKK-1水平(P<0.01),对E-Cadherin无显著影响。结论:刺山柑总生物碱对SSc组织纤维化有一定改善作用,其作用可能与抑制Wnt通路相关。 Objective:To explore the effects of capparis spinosa total alkaloid on Wnt pathways related protein Wnt10 b,Dickkopf-1(DKK-1),E-Cadherin expressions in mice with systemic sclerosis(SSc).Methods:Subcutaneous injection of bleomycin was injected in BALB/c mice to replicate the SSc model.Capparis spinosa total alkaloid cream(225,450,900 mg/kg)was topically applied to the mice daily for 8 weeks.The skin of the injection area of mice was taken,theα-Smooth muscle actin(α-SMA)expression was detected by Western blot,the levels of Wnt10 b and E-Cadherin in skin tissue and Dkk-1 in serum were measured by ELISA.Results:Theα-SMA and Wnt10 b expressions were markedly decreased and the level Dkk-1 was markedly increased after administration of 225,450,900 mg/kg capparis spinosa total alkaloid than those in model group mice(P<0.01).The level of E-Cadherin was not influenced.Conclusion:Capparis spinosa total alkaloids has a certain improvement effect on SSc tissue fibrosis,which may be related to the inhibition of Wnt pathway.
