变应性鼻炎病证结合模型的构建与评价 被引量:8

Establishment and evaluation of the combination with disease and syndrome model of allergic rhinitis

摘要 目的:通过多因素复合建立变应性鼻炎(AR)病证结合动物模型,有利于AR基础研究推进及中医新药制剂研发。方法:冷水负重竭力游泳、高脂饥饿饲养、皮下注射肾上腺素复合卵清蛋白(OVA)致敏的方法制作AR大鼠病证结合动物模型,通过大鼠行为学、鼻黏膜组织病理改变、血流变学、血清IL-4、IFN-γ表达验证病证结合模型。结果:与空白组比较,模型组鼻黏膜上皮破坏,上皮细胞变性增生,黏膜下水肿变性;全血黏度、血浆黏度和红细胞沉降率及IL-4表达升高,IFN-γ表达降低(P<0.05,P<0.01)。中药组治疗后相应各指标较模型组显著改善(P<0.05,P<0.01)。结论:采用上述复合方法制作AR大鼠病证结合模型,结合宏观表征和实验室指标综合评价符合气虚血瘀证AR病证表现,该方法可成功建立AR病证结合动物模型,具有可行性。 Objective:The establishment of animal model of allergic rhinitis(AR)through a multi-factor combination is beneficial to the promotion of basic research of AR and the development of new preparations of traditional Chinese medicine.methods:The AR animal model was induced by cold water weight-bearing swimming,high-fat starvation feeding,subcutaneous injection of epinephrine combined with ovalbumin(OVA).The model was verified by behavioral study,histopathology,hemorheology and the expression of serum IL-4,IFN-γ.Results:Compared with blank group,the nasal mucosa epithelium in the model group was necrotic,the epithelial cells were proliferated,and the mucosal edema was denatured,the whole blood viscosity,high shear reductive viscosity,plasma viscosity,erythrocyte sedimentation rate,IL-4 were significantly increased,and the expression of IFN-γwas decreased(P<0.05,P<0.01).The laboratory indexes improved obviously after treatment in the Chinese herb group than the model group(P<0.05,P<0.01).Conclusion:Combined with macroscopic characterization and laboratory indexes,the AR model of qi deficiency and blood stasis syndrome was comprehensively evaluated.The combined method can successfully establish the animal model of AR and is feasible.
