音乐运动疗法对老年小鼠海马细胞凋亡的影响 被引量:1

Effects of musicokinetic treatment on hippocampal cells apoptosis in geriatric mice

摘要 目的:探究音乐运动疗法对老年小鼠海马细胞凋亡的影响,为防治阿尔茨海默症和帕金森等老年病提供借鉴。方法:将昆明种10月龄雄性老年小鼠32只随机分配入对照组(A组)、有氧运动组(B组)、有氧运动+音乐组(C组)和音乐组(D组),每组8只。运动方式为游泳,40min/次,每周5次。实验8周后,采用免疫组化法观察小鼠海马组织凋亡相关蛋白Bax、Bcl-2、Caspase-3的影响。应用统计软件对海马凋亡细胞进行计数,绘图软件制图观察凋亡细胞。结果:与A组比较,B组、C组和D组Bcl-2表达显著升高(P<0.01),而Bax和Caspase-3表达显著下降(P<0.05,P<0.01);与B组比较,C组和D组Bcl-2表达显著升高(P<0.01),而Bax和Caspase-3显著下降(P<0.05,P<0.01)。与A组和B组比较,C组和D组细胞凋亡数量显著减少(P<0.01)。结论:音乐运动疗法有助于减少老年小鼠海马细胞的凋亡,对神经退行性疾病阿尔茨海默病和帕金森等的预防有一定的借鉴作用。 Objective:The purpose of this study is to explore the effect of musicokinetic therapy on the apoptosis of hippocampal cells in geriatric mice,so as to shed a new light on the prevention and treatment of common Senile Diseases,such as Alzheimer’s disease(AD)and Parkinson’s disease(PD).Methods:A total of 32 geriatric mice,10-month old,were equally randomized into four groups:group A(control group),group B(aerobic exercise group),group C(aerobic exercise+music group)and group D(music group),8 in each group.In this study,exercise mode was swimming,five times for each week.After 8 weeks of experiment,the effects of bax,Bcl-2 and Caspase-3 on apoptosis-related proteins in hippocampus of mice were observed by immunohistochemistry.The apoptotic cells in hippocampus were counted by statistical software,and the apoptotic cells were observed by drawing software.Results:Compared with group A,the expression of Bcl-2 in group B,group C and group D increased significantly(P<0.01),the expression of Bax and Caspase-3 decreased significantly(P<0.05,P<0.01).Compared with group B,the expression of Bcl-2 in group C and group D increased significantly(P<0.01),and the expression of Bax and Caspase-3 decreased significantly(P<0.05,P<0.01).Compared with group A and group B,the number of apoptotic cells in group C and D decreased significantly(P<0.01).Conclusion:Musicokinetic therapy can help reduce the apoptosis of hippocampal cells in geriatric mice,and maybe used for reference in the prevention of neurodegenerative diseases such as AD and PD.
