摘要
为研究lncRNA对鸡骨骼肌生长的影响,在UCSC上预测lncRNA-FKBP1C顺式调控的靶基因为DNMT3B,随后采用qPCR检测DNMT3B在隐性白羽洛克鸡和杏花鸡中的表达量,并在鸡原代成肌细胞中过表达和干扰lnc RNA-FKBP1C,同时对IGF1R基因启动子活性和甲基化进行分析。结果显示,DNMT3B在杏花鸡中差异上调;在鸡原代成肌细胞中发现lncRNA-FKBP1C能抑制DNMT3B、促进IGF1R、FGF12、FGF14、FGF18、FGFR2基因的表达;而DNMT3B可能通过影响IGF1R启动子活性区域的非CpG甲基化来抑制其表达。研究结果提示lncRNA-FKBP1C可能通过顺式调控作用抑制DNMT3B基因的表达,从而影响鸡生长相关基因的表达。
In order to study the effect of lncRNA-FKBP1C on the skeletal muscle development, the target gene of lncRNA-FKBP1C was predicted on UCSC as DNMT3B, and then the expression of DNMT3B was detected between White Recessive Rock(WRR) and Xinghua(XH) chicken by qPCR. LncRNA-FKBP1C was overexpressed and it′s expression was interfered in chicken primary myoblasts, IGF1R gene′s promoter activity and methylation was analyzed. The results showed that the expression of DNMT3B was up-regulated in XH chicken. LncRNA-FKBP1C could inhibit the expression of DNMT3B while promoted the expression of IGF1R, FGF12, FGF14, FGF18 and FGFR2. DNMT3B might inhibit the expression of IGF1R by affecting the non-CpG methylation of its active region. The results suggested that lncRNA-FKBP1 Cmay inhibit the expression of DNMT3B gene through cis-regulation, thereby affecting the expression of chicken growth-related genes.
