从“感觉神经肽P物质-肥大细胞”联动变化探究穴位埋线治疗变应性鼻炎的机制 被引量:5

摘要 目的探讨揭示神经肽P物质(SP)与肥大细胞(MC)的联动变化关系,阐释穴位埋线调节变应性鼻炎(AR)神经源性炎症的机制。方法选取小鼠肥大细胞P815(MCp815)行体外培养,采用SP进行浓度升阶梯干预,初始质量浓度选取0.1 nmol/L,以3为基准倍数,逐渐升高干预质量浓度即0.3、0.9、1968.3 nmol/L。每种干预质量浓度同时设置3个培养基,采用CCK-8法检测MCp815增殖倍数,计算3个培养基的增殖倍数均值。结果SP干预质量浓度与MCp815增殖倍数呈正相关,即SP质量浓度越高,MCp815增殖倍数均值越大,且在低倍质量浓度(0.1、0.3、0.9 nmol/L)时增殖速度最快,进入2.7 nmol/L质量浓度后曲线上升率变缓;高倍质量浓度虽也呈现上升态势,但从218.7 nmol/L开始,曲线逐渐进入平台期,656.1 nmol/L达到增殖巅峰,即使质量浓度再次增加,增殖倍数也不会随之变大。结论SP可以促进MCp815增殖,二者为正相关关系,但在SP质量浓度达到656.1 nmol/L时,达到平台期。研究进一步表明穴位埋线可通过抑制SP进一步下调MC的增殖,从而减轻鼻腔黏膜炎性反应,最终实现治疗作用。 Objective To clarify the mechanism of the acupoint catgut embedding therapy for allergic rhinitis by investigating the linkage relationship between neuropeptide substance P(SP)and mast cells(MC).Methods The mouse mast cell P815(MCp815)was cultured in vitro and then stimulated by SP with the rising concentration.The concentration was initially 0.1 nmol/L and it was gradually increased to 0.3,0.9,1968.3 nmol/L with 3 times as the reference multiple.Three culture media were set simultaneously for each intervention concentration,and the proliferation multiples of MCp815 were detected by CCK-8 and the mean proliferation multiples of the three culture media were calculated.Results The results indicated that the SP intervention concentration was positively correlated with the proliferation multiple of MCp815.The mean proliferation multiple increased with the increase of the SP concentration.The proliferation rates were faster at the concentration of 0.1,0.3 and 0.9 nmol/L,and the rising rate of the proliferation curve slowed down when the concentration reached to 2.7 nmol/L.The proliferation curve showed a rising trend at the high concentration,but the curve gradually entered the plateau stage when the concentration reached to 218.7nmol/L and reached the peak at the concentration of 656.1 nmol/L.Then the proliferation multiple did not increased with the increase of the concentration.Conclusion SP could promote the proliferation of MCp815 with a positive correlation.The positive correlation between SP and MCp815 reaches a plateau stage when the SP concentration reaches 656.1 nmol/L.It is showed in the study that the acupoint catgut embedding therapy could further down-regulate the proliferation of MC by inhibiting SP so as to reduce the inflammatory response of nasal mucosa and finally achieve the therapeutic effect.
