西红花对D-半乳糖致衰老大鼠模型抗氧化能力的影响 被引量:12

摘要 目的研究西红花对D-半乳糖致衰老大鼠模型的抗氧化能力,并探讨其作用机制。方法 SD大鼠随机分为正常对照组、模型对照组、维生素E组和西红花组;建立D-半乳糖诱导的衰老大鼠模型,在皮下注射D-半乳糖15 d后,维生素E组每日上午灌服Vit E;西红花组每日上午灌服西红花;正常对照组、模型对照组从造模第1天开始,分别每日上午灌服等体积的生理盐水。每天观察各组大鼠活动时间、活跃程度、进食量、毛色、精神状态、应激反应、二便等变化;每周称量各组大鼠的体质量;8周后,测定各组大鼠血清以及全脑、肝脏和肾脏匀浆中的超氧化物歧化酶(SOD)活性、谷胱甘肽过氧化物酶(GSH-PX)和丙二醛(MDA)含量。结果与正常对照组相比,模型对照组体质量明显下降,差异有统计学意义(P<0.01);血清以及全脑、肝、肾匀浆中SOD、GSH-Px水平明显下降,而MDA水平显著上升;药物干预组与正常对照组相比,体质量无明显差异(P>0.05),与模型对照组相比,显著增高血清以及全脑、肝脏和肾脏匀浆中的SOD和GSH-Px含量,并显著降低MDA含量。结论西红花能有效提高D-半乳糖致衰老大鼠模型抗氧化能力而达到延缓衰老的作用。 Objective To study the antioxidative ability of saffron to D-galactose-induced aging rat model and explore its mechanism of action. Methods SD rats were randomly divided into normal control group, model control group, vitamin-E group and saffron group. D-galactose-induced aging rat model was established through subcutaneous injection of D-galactose. After 15 days, vitamin-E group was drenched with vitamin-E daily in the morning. The saffron group was drenched with saffron daily in the morning. The normal control group and model control group were drenched with normal saline of equal volume every morning. Daily observations of changes in activity time, activity level, food intake, hair color, mental state, stress response, stool, etc. of each group of rats were performed. The body weight of each group’s rats was weighed weekly. 8 weeks later, we measured superoxide dismutase(SOD) activity, glutathione peroxidase(GSH-PX), and malondialdehyde(MDA) in serum and whole brain, liver and kidney homogenates in each groups. Results Compared with those of the normal control group, the body weight of the model control group was significantly decreased, and there was a significant difference(P<0.01). Levels of SOD and GSH-PX in serum and whole brain, liver and kidney homogenates decreased significantly, while MDA levels increased significantly. Compared with those of the normal control group, the body weights of the vitamin-E and saffron groups were not significantly different(P>0.05). Levels of SOD and GSH-PX increased significantly in serum and whole brain, liver and kidney homogenates, and MDA level reduced significantly. Conclusion Saffron can effectively improve the antioxidant capacity of D-galactose-induced aging rat model and delay the aging.
