摘要 目的确定白屈菜中6种生物碱成分含量的测定方法。方法采用酸提取碱沉淀的方法制备样品,采用HPLC法,条件为1%三乙胺(用磷酸调节pH值至3.0)-乙腈;梯度洗脱,检测波长为274 nm。结果酸提取碱沉淀方法所制的样品杂质少,影响少。HPLC法所建立的梯度洗脱条件,原阿片碱、白屈菜碱、盐酸黄连碱、血根碱、小檗碱和白屈菜红碱分离度较好,精密度、重复性、稳定性和加样回收等试验均符合相关规定。结论所建立的酸提碱沉样品处理法和HPLC含量测定方法,简单易行,可用于白屈菜样品的含量测定,按此方法所测21个样品均检出白屈菜红碱等6种生物碱成分,采自辽宁丹东的白屈菜,白屈菜红碱和总碱含量均较高,质量较好。 Objective To determine the contents of 6 alkaloids in Chelidonium majus. Method The sample was prepared by acid extraction and alkali precipitation. HPLC method was adopted. The condition was 1% triethylamine(adjusting pH to 3.0 by phosphoric acid)-acetonitrile and gradient elution. The detection wavelength was 274 nm. Result The method of acid extraction and alkali precipitation had less impurity and less influence. The gradient elution conditions established by HPLC method showed that the separation of protopine, chelidonine, coptisine hydrochloride, sanguinarine, berberine and chelerythrine was good. Conclusion The treatment method of acid alkali sink sample and HPLC determination method are simple and can be used for content determination of Chelidonium majus sample. Measured by this method,21 samples of Chelidonium majus had chelerythrine base and the contents of chelidonine and total alkaloids of Chelidonium majus harvested from Dandong were high and the quality was good.
