HUC-MSCs对视网膜挫伤大鼠的治疗作用

摘要 目的通过在视网膜挫伤大鼠玻璃体腔内注射体外培养的人脐带间充质干细胞(HUC-MSCs)观察其治疗作用。方法采用组织块法分离纯化HUC-MSCs。自由落体法用于制作大鼠视网膜挫伤模型。44只SD大鼠随机分为正常对照组(N组),挫伤加PBS对照组(C/R+P组)和挫伤加HUC-MSCs治疗组(C/R+M组)。C/R+M组大鼠在挫伤后即刻玻璃体腔内注入P3代HUC-MSCs悬液2μl(2×104/μl),C/R+P组在挫伤后即刻玻璃体腔内注入无菌PBS 2μl。HE染色后使用计算机图像分析系统测量各组大鼠视网膜内外核层的厚度(μm),TUNEL法测量感光细胞的凋亡指数(%)。结果挫伤后1h,1d,C/R+P组视网膜外核层平均厚度大于C/R+M组,挫伤后7d,14d,C/R+P组视网膜外核层平均厚度小于C/R+M组,差异均有显著性(P<0.05)。挫伤后3d,7d,14d,C/R+P组视网膜感光细胞凋亡指数大于C/R+M组,差异有显著性(P<0.05)。结论视网膜挫伤能导致视网膜感光细胞凋亡,玻璃体腔注射HUC-MSCs可抑制视网膜挫伤大鼠感光细胞的凋亡,保护视网膜结构。 Objective Observing the therapeutic effect of injecting Human umbilical cord mesenchymal stem cells(HUC-MSCs) in vitro culture of rat with retinal contusion.Methods Purified HUC-MSCs were isolated by tissue block method,and free fall method was used to make a model of retinal bruising in rats.Forty-four SD rats were randomly divided into normal control group(N group),contusion plus PBS control group(C/R+P group) and contusion plus HUC-MSCs treatment group(C/R+M group),the C/R+M group was injected with P3 HUC-MSCs suspension 2μl(2×10~4/μl) into the vitreous cavity immediately after the injury model was made,and the C/R+P group was injected with PBS 2μl into the vitreous cavity.After HE staining,measuring the thickness of inner and outer retinal layers under computer-based image analysis system,TUNEL technique was used to assess apoptosis index(AI).Results HUC-MSCs transplanted treatment results observed:after 1 h,1 d,the retinal outer nuclear layer average thickness of C/R+M group was thicker than C/R+P group(P<0.05);After 3 d,7 d,14 d,C/R+P group retinal photosensitive apoptosis index greater than C/R+M group,the difference was statistically significant(P<0.05). Conclusion Retinal contusion leads to apoptosis of photoreceptor cells,injection of HUC-MSCs into vitreous cavity can inhibit the apoptosis of photoreceptor cells in rats with retinal contusion and protect retinal structure.
