摘要
建立了小花清风藤茎叶中两种五环三萜类化合物,24-羟基-3-氧代-12-齐墩果烯-28-酸(化合物1)与齐墩果酸(化合物2)的质量分数测定方法.采用高效液相色谱法分别测定小花清风藤茎叶中化合物1和化合物2的质量分数.色谱条件为:Venusil XBP C18(5μm,250×4.6mm)色谱柱,恒定流速为1.0mL/min,设检测波长为203nm,柱温30℃.化合物1的流动相条件为:乙腈-水(90∶10),化合物2的流动相条件为:甲醇-水(83∶17).结果表明:小花清风藤茎叶中化合物1的质量分数为0.09213mg/g,化合物2质量分数为0.3471mg/g.化合物1在0.2575~1.0300mg/mL范围内与色谱峰面积呈良好的线性关系,r=0.9997,平均回收率为95.56%,RSD(n=5)为1.15%.化合物2进样量在0.4000~0.8000mg/mL范围内与峰面积呈良好的线性关系,r=0.9996,平均回收率为97.56%,RSD(n=5)为1.06%.该方法简便准确,重现性好,可以作为小花清风藤茎叶中两种三萜类成分化合物1和化合物2的质量分数测定方法.
A high-performance liquid chromatography(HPLC)method for the content determination of 24-hydroxy-3-oxoolean-12-en-28-oic acid(compound 1)and oleanolic acid(compound 2)in stems and leaves of Sabia Parviflora was established.The HPLC was used to determine the contents of compound 1 and compound 2 in stems and leaves of Sabia Parviflora.The method was carried out on a Venusil XBP C18 chromatographic column(5μm,250 mm×4.6 mm),the flow rate was 1.0mL/min with a detection wavelength of 203 nm and the column temperature was kept at 30℃.The mobile phases of compound 1 and compound 2 were acetonitrile-water(90∶10)and methanol-water(83∶17),respectively.The contents of compound 1 and compound 2 in stems and leaves of Sabia Parviflora had been determined as 0.09213 mg/g and 0.3471 mg/g,respectively.The calibration curve of compound 1 showed a good linear relationship at the range of 0.2575-1.0300 mg/mL with its linear coefficient of 0.9997;and the average recovery was determined as 95.56% with RSD(n=5)1.15%.The calibration curve of compound 2 showed a good linear relationship at the range of 0.4000-0.8000 mg/mL with its linear coefficient of 0.9996;and the average recovery was determined as 97.56% with RSD(n=5)1.06%.The proposed method is simple,accurate and repeatable that can be used for quality control of compound 1 and compound 2 in stems and leaves of in Sabia Parviflora.
