活血止痛方调控Wnt/β-catenin信号通路对膝骨性关节炎的影响 被引量:1

摘要 目的探讨Wnt/β-catenin信号通路在滕骨关节炎发病中的作用,阐述活血止痛方干预膝骨性关节炎的作用机制。方法将36只雄性SD大鼠随机分为中药组、模型组和对照组。中药组和模型组复制膝关节骨关节炎模型,中药组术后第5周开始灌服活血止痛方汤药,福型组和对照组灌服等量的生理盐水,分别于术后第8、12周分批处死大鼠,于膝关节滑膜组织及软骨组织取材,行病理形态学观察、Mankin评分评估;检测软骨组织中β-catenin.MMP-13和TNF-a基因的表达水平。结果中药组和模型组膝关节滑膜增生、水肿及软骨磨损性变随时间延长而加重,以模型组更明显;不同时间点Mankin评分,模型组均高于中药组和对照组;软骨组织中β-catenin,MMP-13和TNF-a基因mRNA表达量与Mankin评分结果相似。结论活血止痛方能有效保护膝关节软骨,从而延缓骨关节炎进程。其作用机制可能与其抑制β-catenin的过度表达,从而阻断Wnt/β-catenin信号通路表达有关。 Objective To investigate the role of Wnt/β-catenin signaling pathway in the pathogenesis of knee osteoarthritis to explore the mechanism of Huoxue Zhitong Prescription on intervention of knee osteoarthritis.Methods All 36 male SD rats were randomly divided into three subgroups:Chinese herbal medicine group(A subgroup),model group(B subgroup)and sham group(C subgroup).The rats were sarificed in batches at 8 and 12 weeks after surgery,and the following the parameters of synovial and cartilage tssue of the knee joint were observed and measured,including pathomorphological observation,Mankin score and expression levels of beta-catenin,MMP-13 and TNF-alpha genes.Result The synovial hyperplasia,edema and cartilage abrasion of the A subgroup and the B subgroup worsened gradually with the prolongation of observation time,especially in the B subgroup.The Mankin score of the B subgroup was higher than that of the A subgroup and the C subgroup at diferent time points.The expresion levels of beta-catenin,MMP-13 and TNF-alpha genes of the B subgroup was higher than that of the A subgroup and the C subgroup in cartilage tssue at different time points.Conclusion Huoxue Zhitong Prescription can efectively prevent cartilage abrasion to delay the progress of knee osteoarthritis.Is mechanism may be related with blocking the expression of W nt/beta-catenin signaling pathway by inhibiting the overexpression of beta-catenin.
