摘要
目的观察微小RNA(miRNA,miR)-140在软骨细胞中的表达并探讨其临床意义。方法收集股骨头32例,按照软骨退变情况,分为骨关节炎(OA)组17例,对照组15例。同时收集两组患者临床资料及相关生化指标,采用实时荧光定量聚合酶链式反应检测两组的miR-140的表达水平,比较两组的临床资料、生化指标和miR-140的表达水平,同时结合相关指标,分析miR-140与其相关性,评估影响miR-140的因素及临床意义。结果两组患者白细胞、血红蛋白、白蛋白、尿酸、血沉及C-反应蛋白差异无统计学意义,OA组的体重指数(BMI)高于对照组,差异有统计学意义(t=2.436,P<0.05)。miR-140在OA组的相对表达水平为(0.14±0.03),低于对照组(1.02±0.23),差异有统计学意义(t=-14.892,P<0.05)。相关性分析显示,miR-140与年龄、BMI呈正相关(r=0.852、0.809,P<0.05)。结论miR-140在骨关节炎患者中表达下降并与体重指数及年龄因素明显相关。
Objective To observe the expression and clinical implication of microRNA(miRNA,miR)-140 in chondrocytes.Methods 32 femoral heads were divided into OA group(17 cases)and control group(15 cases).At the same time,we collected the clinical data and related biochemical parameters of the two groups,detected the expression level of miR-140 by real-time fluorescence quantitative polymerase chain reaction,combined the clinical data and laboratory indexes of the two groups with the expression level of miR-140,analyzed the correlation,and evaluated the relevant factors and clinical significance which affect miR-140.Results There were no significant difference between the two groups in terms of leukocyte,hemoglobin,albumin,uric acid,ESR and C-reactive protein.The BMI of osteoarthritis group was significantly higher than that of the control group(t=2.436,P<0.05).The relative expression level of miR-140 in OA group(0.14±0.03),in the control group(1.02±0.23),was lower than that in the control group(t=-14.892,P<0.05).miR-140 was positively correlated with age and BMI(r=0.852,0.809,P<0.05).Conclusion The expression of miR-140 decreased in osteoarthritis patients and was related to BMI and age.
