补骨脂素联合骨形成蛋白2对骨折愈合的影响 被引量:1

Effect of psoralen combined with bone morphogenetic protein 2 on fracture healing

摘要 目的观察补骨脂素联合重组人骨形态发生蛋白2(rhBMP-2)对山羊股骨骨折愈合和生物力学的影响。方法将24只20~30月龄健康山羊(来源于山西医科大学第一医院)采用完全随机法分为为模型组(灌服1 ml 0.9%生理盐水)、补骨脂素组[灌服8 mg/(kg·d)补骨脂素溶液],rhBMP-2组[6 g/(kg·d)rhBMP-2灌服],补骨脂素联合rhBMP-2组[灌服rhBMP-26 g/(kg·d)与补骨脂素8 mg/(kg·d)]。术后120 d处死动物,X线片检测骨愈合;游标卡尺测量骨皮质厚度和骨髓腔直径;858 Mini Bionix型材料测试系统测试股骨最大载荷和弹性载荷。组间比较采用LSD-t检验法进行组间两两分析。结果补骨脂素+rhBMP-2组的骨愈合度、骨髓腔直径、股骨最大载荷、弹性荷载分别为(89.14±6.81)mm3、(26.23±3.11)mm、(387.78±56.36)N、(106.23±22.11)Mpa均高于模型组[(63.47±7.16)mm3、(13.41±3.12)mm、(152.32±22.36)N、(55.63±12.18)Mpa],差异有统计学意义(t=12.115、9.158、10.232、12.523,P<0.05);补骨脂素+rhBMP-2组的上述指标高于补骨脂素组[(71.52±8.44)mm3、(17.27±1.03)mm、(221.71±43.47)N、(58.45±18.27)Mpa],差异均有统计学意义(t=7.521、6.332、8.201、6.511,P<0.05);补骨脂素+rhBMP-2组的上述指标高于rhBMP-2组[(68.14±5.28)mm3、(19.12±2.51)mm、(238.45±31.52)N、(61.04±17.96)Mpa],差异有统计学意义(t=5.661、7.052、6.044、5.206,P<0.05)。另外,补骨脂素+rhBMP-2组的骨皮质厚度[(1.12±0.10)mm]低于模型组[(2.11±0.09)mm]、补骨脂素组[(1.45±0.15)mm]、rhBMP-2组[(1.32±0.08)mm],差异均有统计学意义(t=15.358、8.551、7.141,P<0.05)。结论补骨脂素联合rhBMP-2是潜在的股骨骨折愈合关键药物之一。 Objective To investigate the effects of psoralen combined with recombinant human bone morphogenetic protein 2(rhBMP-2)on the healing and biomechanics of goat femoral fractures.Methods Twenty-four healthy goats(from the first hospital of Shanxi Medical University)aged 20 to 30 months were divided into model group(infused with 1 ml 0.9%physiological saline),psoralen group[infused with 8 mg/(kg·d)psoralen solution],rhBMP-2 Group[6 g/(kg·d)rhBMP-2],psoralen combined with rhBMP-2 group[rhBMP-26 g/(kg·d)and psoralen 8 mg/(kg·d)]by completely random method.The animals were sacrificed 120 days after operation,and bone healing was detected by X-ray films;Vernier calipers measure the thickness of the bone cortex and the diameter of the bone marrow cavity;The 858 Mini Bionix material testing system tests the max-load and elasticity load.The LSD-t test method was used for comparison between groups to conduct pairwise analysis between groups.Results The bone healing degree(mm3),bone marrow cavity diameter(mm),maximum femoral load(N),and elastic load(Mpa)of each group were compared.The above indicators of the psoralen+rhBMP-2 group(89.14±6.81,26.23±3.11,387.78±56.36,106.23±22.11)were higher than the model group(63.47±7.16,13.41±3.12,152.32±22.36,55.63±12.18),the difference was statistically significant(t=12.115,9.158,10.232,12.523,P<0.05);the above indicators of the psoralen+rhBMP-2 group were higher than those of the psoralen group(71.52±8.44,17.27±1.03,221.71±43.47,58.45±18.27),the differences were statistically significant(t=7.521,6.332,8.201,6.511,P<0.05);psoralen+rhBMP-2 The above indicators of the rhBMP-2 group were higher than those of the rhBMP-2 group(68.14±5.28,19.12±2.51,238.45±31.52,61.04±17.96),and the difference was statistically significant(t=5.661,7.052,6.044,5.206,P<0.05).In addition,the thickness of cortical bone[(1.12±0.10)mm]of the psoralen+rhBMP-2 group was lower than that of the model group(2.11±0.09 mm),the psoralen group[(1.45±0.15)mm],and the rhBMP-2 group[(1.32±0.08)mm],the differences were statistically significant(t=15.358,8.551,7.141,P<0.05).Conclusion Psoralen combined with rhBMP-2 is one of the potential drugs for the healing of femoral fractures,providing a basis for basic research on femoral fractures.
