环状RNA hsacirc0007637在非小细胞肺癌组织的表达及其临床意义 被引量:3

Expression and clinical significance of circular RNA hsa_circ_0007637 in non-small cell lung cancer

摘要 目的观察hsacirc0007637在非小细胞肺癌(NSCLC)组织中的表达,并探讨其临床意义。方法收集2017年2月至2018年9月从广西医科大学第一附属医院NSCLC患者的癌组织和癌旁组织样本25对,提取组织标本RNA,采用实时定量反转录聚合酶链反应(RT-qPCR)法检测hsacirc0007637在NSCLC及癌旁组织中的表达,并分析其与临床病理特征的相关性,受试者工作特征曲线(ROC)评估hsacirc0007637的诊断价值。组间比较采用t检验以及单因素方差分析。结果hsacirc0007637在NSCLC组织和肺癌细胞株中的表达水平显著低于相邻的正常组织和细胞(t=2.791,P<0.01);hsacirc0007637在癌组织中的低表达与肿瘤直径及淋巴结转移明显相关(t=2.385,P<0.05;t=2.296,P<0.05)。分析ROC曲线得知,hsacirc0007637诊断NSCLC的曲线下的面积(AUC)为0.855,灵敏度为92.3%,特异度为73.1%(P<0.01)。结论hsacirc0007637在NSCLC组织和细胞中低表达,并与肿瘤大小和淋巴结转移密切相关。 Objective To observe the expression of hsa_circ_0007637 in non-small cell lung cancer(NSCLC)tissues and explore its clinical significance.Methods Twenty-five pairs of cancer tissue and paracancer tissue samples of postoperative NSCLC patients were collected and sample RNA was extracted.The expression of hsa_circ_0007637 in NSCLC and adjacent tissues was detected by real-time quantitative reverse transcriptase-polymerase chain reaction(RT-qPCR),and its correlation with clinicopathological features was analyzed.The receiver operating characteristic(ROC)of the subjects was used to assess the diagnostic value of hsa_circ_0007637.Results The expression level of hsa_circ_0007637 in NSCLC tissues and lung cancer cell lines was significantly lower than that in adjacent normal tissues and cells(t=2.791,P<0.01).The low expression of hsa_circ_0007637 in cancer tissues was obviously correlated with tumor diameter and lymph node metastasis(t=2.385,P<0.05;t=2.296,P<0.05).ROC curve analysis showed that the area under curve(AUC)of NSCLC diagnosed by hsa_circ_0007637 was 0.855,the sensitivity was 92.3%,and the specificity was 73.1%(P<0.01).Conclusion hsa_circ_0007637 is low expressed in NSCLC tissues and cells,and is closely related to tumor size and lymph node metastasis.
