萎平舒胶囊对慢性萎缩性胃炎模型大鼠胃黏膜细胞自噬水平的影响

Effect of Weipingshu Capsule on the Autophagy Level of Gastric Mucosal Cellsin in Chronic Atrophic Gastritis Model Rats

目的探讨萎平舒胶囊对慢性萎缩性胃炎伴异型增生模型大鼠胃黏膜细胞自噬水平的影响。方法60只Wistar大鼠中,随机选取8只作为正常对照组,其余以MNNG诱导法建立大鼠胃粘膜萎缩伴肠化的模型。随机分为模型组、萎平舒低剂量组、萎平舒高剂量组,分别观察大鼠胃黏膜组织病理学变化、胃窦胃体黏膜与肌层厚度变化,各组大鼠胃黏膜上皮细胞中自噬小体的变化。结果与模型组比较,萎平舒胶囊低、高剂量组窦部的胃粘膜厚度及肌层厚度与模型组比较显著增厚(P<0.01;P<0.01),萎平舒大剂量组细胞质内可见到较多的次级溶酶体,自噬样小体数量较模型组较多。结论萎平舒胶囊能明显改善胃粘膜上皮细胞形态异常及腺体萎缩,可使胃黏膜上皮细胞内自噬小体数量增多。

Objective To investigate the effect of Weipingshu(soothing stomach)Capsule on the autophagy level of gastric mucosal cells in chronic atrophic gastritis rat model associated with dysplasia.Methods 8 were randomly selected as the normal control group among 60 Wistar rats,and the rest were induced by MNNG to establish a model of gastric mucosal atrophy with intestinal metaplasia,and randomly divided into model group,Weipingshu low-dose group and Weipingshu high-dose group.The histopathological changes of gastric mucosa,gastric antrum and gastric body mucosa and muscle layer thickness as well as the gastric mucosal epithelial cells changes in autophagosomes of each group were observed respectively.Results Compared with the model group,the thickness of gastric mucosa and muscle layer in the antrum of the Weipingshu capsule low and high dose groups was significantly higher than that of the model group(P<0.01;P<0.01).The secondary?lysosome in the Weipingshu high dose groupvisible is visible with more autophagy-like bodies.Conclusion Weipingshu Capsule can significantly improve the abnormal morphology of gastric mucosal epithelial cells and gland atrophy,increasing the number of autophagosomes in gastric mucosal epithelial cells.
