自拟益智方结合音乐治疗对阿尔茨海默病模型小鼠的干预研究 被引量:1

摘要 目的探讨自拟益智方结合音乐治疗在行为学实验、脑内生化指标及脑组织病理学方面对阿尔茨海默病(AD)模型小鼠的干预影响。方法将普通级雌性NIH小鼠随机分为5组:对照组、AD模型组(简称模型组)、自拟益智方(26 mg·kg-1·d-1)组、自拟益智方结合音乐组(简称结合组)及多奈哌齐(0. 75 mg·kg-1·d-1)组,每组15只。结合组每天选择轻快、舒缓类型音乐重复播放1h,模型组和给药各组腹腔注射D-半乳糖120 mg·kg-1·d-1和亚硝酸钠90 mg·kg-1·d-1,连续60 d,对照组腹腔注射等量的生理氯化钠溶液。从第47天开始,给药各组灌胃益智方和多奈哌齐,连续14 d,对照组和模型组灌胃等量的生理氯化钠溶液。结果与对照组比较,模型组小鼠逃避潜伏期、平台穿越频次、脑组织中SOD、Ach E各指标差异有统计学意义(P <0. 05);与模型组比较,结合组小鼠逃避潜伏期、平台穿越频次、脑组织中SOD、Ach E各指标差异有统计学意义(P <0. 05)。脑组织病理形态观察也显示,结合组小鼠与模型组比较明显改善,大部分区域神经元排列整齐,细胞核清晰,尼氏体较均匀丰富,神经纤维无明显缠结,可见卫星现象。结论自拟益智方结合音乐治疗对D-半乳糖联合亚硝酸钠诱导的AD小鼠有一定神经保护作用,可以改善AD小鼠的学习记忆能力,产生抗AD的药理作用。其作用机制可能是通过提高小鼠脑组织中SOD活性,提高机体的抗氧化及自由基的清除能力;同时降低Ach E活力,从而提升脑内胆碱能系统功能有关。 Objective To explore the effects of self-made Yizhi recipe combined with music therapy on behavior experiment,brain biochemical index and brain histopathology in mice with Alzheimer’s disease( AD). Methods Normal female NIH mice were randomly divided into five groups: control,AD model( abbreviated as model),self-made Yizhi recipe( 26 mg·kg-1·d-1),self-made Yizhi recipe combined with music( abbreviated as combination) and donepezil( 0. 75 mg·kg-1·d-1),with 15 mice in each group. In the combination group,light and soothing music was repeated for 1 hour every day. D-galactose 120 mg·kg-1·d-1 and sodium nitrite 90 mg·kg-1·d-1 were injected intraperitoneally in the model group and the administered groups for 60 days. The control group was given intraperitoneal injection of the same amount of physiological sodium chloride solution. From the 47 th day,each administered group was given Yizhi recipe or Donepezil for 14 days. The control group and model group were given the same amount of physiological sodium chloride solution. Results There were significant differences in escape latency,frequency of platform crossing,SOD and Ach E in brain tissue between the model group and the control group( P < 0. 05). There were significant differences in escape latency,platform crossing frequency,SOD and Ach E in brain tissue between the combination group and the model group( P <0. 05). Pathological observation of brain tissue also showed that compared with the model group,the mice of the combination group improved significantly. Most of the neurons were arranged neatly,the nucleus was clear,the Nissl bodys were more uniform and abundant,and the nerve fibers were not tangled. Satellite phenomenon could be seen. Conclusion The self-made Yizhi recipe combined with music therapy has neuroprotective effect on AD mice induced by D-galactose and sodium nitrite. It can improve the learning and memory ability of AD mice and produce anti-AD pharmacological effect. Its mechanism may be related to the enhancement of SOD activity in mouse brain tissue,the enhancement of antioxidant and free radical scavenging capacity,and the reduction of Ach E activity,thereby enhancing the function of cholinergic system in brain.
