摘要
目的研究人参活性成分原人参二醇(Protopanaxdiol,PPD)对非小细胞肺癌的抑制作用及机制。方法将人非小细胞肺癌A549细胞分为对照组(等量二甲基亚砜DMSO)和实验组(30μg·mL-1 PPD)。用JC-1标记的流式细胞术检测细胞线粒体膜电位,用激光共聚焦显微镜观察线粒体通透性转换孔道(mitochondrial permeability transition pore,mPTP)变化,用Western blot实验检测解偶连蛋白-2(Uncoupling protein 2,UCP2)的表达水平。结果实验组和对照组红绿荧光的相对比例分别为0.29±0.05和2.44±0.03,钙黄绿素的荧光强度分别为2.79±0.25和8.58±1.83,UCP2蛋白相对表达量分别为0.44±0.03和1.00±0.15,差异均有统计学意义(均P<0.05)。结论 PPD可诱导人非小细胞肺癌A549细胞凋亡,其机制可能与抑制UCP2的基因表达和调节线粒体功能相关。
Objective To explore the inhibitory effect and mechanism of protopanaxadiol, an active ingredient of ginseng, on non-small cell lung cancer. Methods Non-small cell lung cancer A549 cell line was divided into two groups: control group(equal dimethyl sulfoxide DMSO) and test group(PPD dilution 30 μg·mL-1). JC-1 labeled flow cytometry was used to detect cell mitochondrial membrane potential, laser confocal microscope was used to observe the changes of mitochondrial permeability transition pore(mPTP), Western blot experiment detected the expression level of uncoupling protein 2(UCP2). Results In the test group and control group, the relative ratio of red-green fluorescence was 0.29±0.05 and 2.44±0.03, the fluorescence intensity of calcein was 2.79±0.25 and 8.58±1.83, the relative expression of UCP2 protein was 0.44±0.03 and 1.00±0.15,the differences of the factors were significant(all P<0.05). Conclusion PPD can inhibit the proliferation and induce apoptosis of human lung adenocarcinoma A549 cells, and its mechanism may be related to inhibiting UCP2 gene expression and regulating mitochondrial function.
