黑米花青苷对糖尿病大鼠视网膜病变的保护作用 被引量:5

Protective effect of black rice anthocyanin on retinopathy in diabetic rats

摘要 目的观察黑米花青苷(AEBR)对糖尿病大鼠视网膜病变的保护作用。方法用腹腔注射链脲佐菌素构建糖尿病模型大鼠,并随机分为模型组(9只)、对照组(7只)和实验组(7只),另取9只正常大鼠作为正常组。正常组和模型组均灌胃给予蒸馏水5 mL·kg^-1·d^-1;对照组灌胃给予二甲双胍200 mg·kg^-1·d^-1;实验组灌胃给予AEBR 200 mg·kg^-1·d^-1。4组大鼠均连续给药8周。比较4组大鼠的血糖、视网膜组织结构及其形态计量学参数、超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量变化。结果给药8周后,实验组、对照组、模型组和正常组的随机血糖分别为(23.50±3.04),(23.06±0.95),(31.88±1.89)和(5.23±0.97)mmol·L^-1,SOD分别为(201.67±5.32),(209.09±34.58),(180.07±21.65)和(198.43±10.85)U·mL^-1,MDA分别为(7.46±0.97),(7.08±1.64),(8.80±0.58)和(4.83±1.07)nmol·mL^-1,视网膜总厚度分别为(116.68±23.72),(110.14±19.35),(93.62±23.50)和(168.82±39.30)μm。实验组和模型组相比,上述指标均得到改善,差异均有统计学意义(P<0.05或P<0.01)。结论 AEBR通过提高机体抗氧化水平,来保护糖尿病大鼠视网膜,延缓视网膜病变。 Objective To observe the protective effect of black rice anthocyanin(AEBR) on retinopathy of diabetic rats. Methods Streptozotocin was injected intraperitoneally to construct diabetic model rats, which were randomly divided into model group(n=9), control group(n=7) and experimental group(n=7), another 9 normal rats were taken as normal group. The normal and model groups were given 5 mL·kg^-1·d^-1of distilled water by gavage;the control group was given metformin 200 mg·kg^-1·d^-1 by gavage;the experimental group was given AEBR 200 mg·kg^-1·d^-1 by gavage. The rats in the four groups were continuously administered for 8 weeks. The changes of blood glucose, retinal tissue structure and morphometric parameters,superoxide dismutase(SOD) activity, and malonaldehyde(MDA) content of the rats in four groups were compared. Results After 8 weeks of administration,the random blood glucose of experimental group,control group,model group and normal group were(23.50±3.04),(23.06±0.95),(31.88±1.89) and(5.23±0.97)mmol·L^-1, SOD levels were(201.67±5.32),(209.09±34.58),(180.07±21.65)and(198. 43 ± 10. 85) U·mL^-1,MDA levels were(7. 46 ± 0. 97),(7. 08 ± 1. 64),(8. 80 ± 0. 58) and(4. 83 ± 1. 07) nmol·mL^-1,the total thickness of the retina were(116. 68 ± 23. 72),(110. 14 ± 19. 35),(93. 62 ± 23. 50) and(168. 82 ± 39. 30) μm. Compared with the model group,the above indicators of the experimental group have been improved,and the differences were significant(P < 0. 05 or P < 0. 01). Conclusion AEBR can protect the retina of diabetic rats and delay the progress of retinopathy by increasing the antioxidant ability of diabetic rats.
