圆齿野鸦椿果皮总三萜的提取工艺及其抗肿瘤活性 被引量:9

摘要 目的:探究圆齿野鸦椿果皮总三萜的提取工艺及其抗肿瘤活性。方法:利用超声提取法进行圆齿野鸦椿果皮总三萜的提取,通过单因素试验和响应面法对提取工艺进行优化,采用噻唑蓝比色法(MTT)检测果皮总三萜对肝癌细胞HepG2、人肺癌细胞A549、乳腺癌细胞MCF-7和结肠癌细胞HCT-116的增值抑制作用。结果:圆齿野鸦椿果皮总三萜提取的最佳工艺条件为提取时间70 min、超声功率400 W、乙醇体积分数67%和液料比35∶1,此条件下的提取率可高达9.63%。圆齿野鸦椿果皮总三萜对HepG2、A549、HCT-116和MCF-7细胞的半数抑制浓度(IC50)分别为(87.56±1.35)、(92.38±0.96)、(89.95±1.64)、(97.30±1.27)μg·mL^-1。结论:采用响应面法获得圆齿野鸦椿果皮总三萜的最佳提取工艺条件,并以总三萜产物对多种肿瘤细胞进行体外抗肿瘤实验,发现圆齿野鸦椿果皮总三萜具有良好的抗肿瘤活性。 Objective:To study the extraction technology and antitumor activity of total triterpenes from the pericarp of Euscaphis konishii Hayata(TTEP).Methods:Ultrasonic extraction method was used to extract TTEP and the extraction process was optimized by single factor test and response surface method.Methyl thiazdyl tetrazolium(MTT)method was adopted to detect the proliferation inhibition of TTEP on liver cancer cell HepG2,lung cancer cell A549,breast cancer cell MCF-7 and colon cancer cell HCT-116.Results:The best extraction yield could reach 9.63%,which was obtained under the condition of 67% ethanol solution,(35∶1)mL·g^-1 liquid-material ratio,70 min extraction time,and 400 W ultrasonic power calculated by the Response surface methodology.The IC50 of TTEP for HepG2,A549,McF-7 and HCT-116 were(87.56±1.35),(92.38±0.96),(89.95±1.64),(97.30±1.27)μg·mL^-1,respectively.Conclusion:The optimum extraction conditions of total triterpenes were obtained by response surface method,and anti-tumor experiments were carried out on a variety of tumor cells with total triterpene products.The total triterpenes from the pericarp of E.konishii has potent anti-cancer ability.
