摘要
目的:研究不同场强电磁脉冲(EMP)对大鼠情绪性行为的影响,为电磁辐射产生的生物效应提供实验基础。方法:将成年雄性SD大鼠随机分为对照组、125 kV/m组和250 kV/m组,每组8只。通过旷场实验与高架十字迷宫实验研究EMP不同场强辐照对大鼠情绪性行为的影响。用ELISA方法检测大鼠大脑皮层与海马组织中的5-HT表达水平。结果:与对照组相比,125 kV/m组和250 kV/m组中央区停留时间、穿越中央区次数有所下降,其中250 kV/m组与对照组比较差异存在统计学意义(P<0.05)。与对照组相比,125 kV/m组和250 kV/m组进入开臂停留时间百分比和进入开臂次数百分比均降低,其中250 kV/m组与对照组比较差异存在统计学意义(P<0.05)。与对照组相比,250 kV/m组大鼠皮层组织5-羟色胺(5-HT)表达水平有所增加(P<0.05),125 kV/m组和250 kV/m组大鼠海马组织5-HT表达水平均升高(均P<0.05)。结论:250 kV/m电磁脉冲连续辐照15 d会诱发大鼠焦虑样行为表现,这种情绪性行为改变可能与大鼠大脑皮层及海马组织的5-HT表达增加有关。
Objective:To study the effects of electromagnetic pulses(EMP)with different field intensity on emotional behaviors in rats,and to provide experimental basis for the biological effects of electromagnetic radiation.Methods:Adult male SD rats were randomly divided into control group,125 kV/m group,and 250kV/m group,with 8 rats in each group.The effects of EMP radiation on emotional behavior of rats were studied by open field test and elevated plus maze test.ELISA method was used to detect 5-HT expression level in rat cerebral cortex and hippocampus.Results:Compared with the control group,the staying time and the number of crossing the central area of rats in the 125 kV/m group and 250 kV/m group decreased,and the 250 kV/m group was significantly different from the control group(P<0.05).Compared with the control group,the proportion of the staying time and the number of times entering the open arm of rats in the 125 kV/m group and the 250 kV/m group both decreased,and the 250 kV/m group was significantly different from the control group(P<0.05).Compared with the control group,the expression level of 5-HT in the cortical tissue of rats in the 250 kV/m group was increased(P<0.05),and the expression levels of 5-HT in the hippocampal tissue of rats in the 125 kV/m group and the 250 kV/m group were also increased(all P<0.05).Conclusion:After EMP irradiation for 15 days with 250 kV/m,the rats showed anxiety-like behavior,and this change of emotional behavior may be related to the increased expression of 5-HT in the cerebral cortex and hippocampus of the rats.
