转录因子12在卵巢癌中的表达及其与预后的相关性

Expression of TCF12 in ovarian cancer and its relationship with prognosis

目的:探讨转录因子12(transcription factor 12,TCF12)在卵巢癌组织中的表达情况,及其与临床病理参数和预后之间的关系。方法:收集卵巢癌患者组织,用实时定量聚合酶链式反应(real-time quantitative polymerase chain reaction,RT-qPCR)检测卵巢癌及其癌旁组织中TCF12的表达情况,并用免疫组织化学的方法检测116例卵巢癌患者中TCF12的表达情况,分析其表达与年龄、病理学类型、FIGO分期、组织学分级、转移之间的关系,用Kaplan-Meier法分析TCF12在卵巢癌患者中的表达高低与生存期之间的关系。结果:RT-qPCR及免疫组织化学结果均显示卵巢癌组织中的TCF12表达量明显高于其对应的癌旁组织(P<0.01);TCF12的表达水平与卵巢癌的组织学分级(P=0.019)、转移(P=0.002)呈正相关;生存曲线分析显示TCF12高表达患者的总生存期明显短于低表达者(P<0.01)。结论:TCF12在卵巢癌患者的肿瘤组织中高表达,提示TCF12可能参与卵巢的发生发展,且TCF12在肿瘤组织中高表达可能会影响卵巢癌的转移,提示TCF12可作为潜在的卵巢癌预后指标。

Objective:To investigate the expression profiles of transcription factor 12(TCF12)in ovarian cancer tissues and its relationship with clinicopathological factors and prognosis.Methods:The clinical tumor tissues of patients with ovarian cancer were collected,and real-time quantitative polymerase chain reaction(RT-qPCR)was used to examine the expression of TCF12 in ovarian cancer tissues and matched non-tumor para-neoplastic tissues.116 cases of pathological specimens from patients undergoing surgery for ovarian cancer in our hospital were also collected and immunohistochemical staining method was used to detect the expression of TCF12.The association between TCF12 expression with age,pathological type,FIGO stage,histological grade or metastasis was further analyzed.Kaplan-Meier survival curve was used to analyze the cumulative overall survival rate.Results:The results showed that TCF12 was high expressed in ovarian tumor tissues compared with that ones in para-neoplastic tissues(P<0.01).High expression of TCF12 was correlated with clinicopathological factors histological grade(P=0.019)and metastasis(P=0.002).Kaplan-Meier survival curve showed that ovarian cancer patients who presented with high TCF12 expression had poor overall survival rates compared with patients who presented with low expression(P<0.01).Conclusion:TCF12 is high expressed in tumor tissues of patients with ovarian cancer,suggests that TCF12 may be involved in the development and progression of ovarian cancer;and high TCF12 expression in tumor tissues may affect the metastasis of ovarian cancer,and suggests that TCF12 may serve as a prognostic factor for ovarian cancer.
