摘要
【目的】探究中国板栗主栽品种‘燕山红栗’胚性愈伤组织对遗传霉素的耐受力。【方法】以‘燕山红栗’(Castanea mollissima Blume‘Yanshanhongli’)幼胚胚尖诱导出的两种不同状态的胚性愈伤组织为材料,在含有不同质量浓度遗传霉素的固体和液体扩繁培养基上培养,观察其生长状况,并统计其死亡率和鲜质量变化量。【结果】在固体培养基上,遗传霉素对两种胚性愈伤组织的致死质量浓度分别为130 mg/L和80 mg/L;液体悬浮培养中,遗传霉素对两种胚性愈伤组织的致死质量浓度分别为60 mg/L和30 mg/L。【结论】确定在固体和液体悬浮培养条件下,两种板栗胚性愈伤组织对遗传霉素的耐受质量浓度,为板栗转基因过程中遗传霉素的使用提供依据。
【Objective】To investigate the tolerance of callus from Chinese chestnut cultivar Castanea mollissima Blume‘Yanshanhongli’to geneticin.【Methods】Two kinds of embryogenic calli induced from the immature embryo of C.mollissima‘Yanshanhongli’were used as materials.The sensitivity of embryogenic callus to geneticin was determined by death rate and variation in fresh weight after inoculated into proliferation medium supplemented with different levels of geneticin.【Results】Under the solid culture,the lethal concentrations of geneticin to these two kinds of embryogenic callus were 130 mg/L and 80 mg/L,respectively.While under the suspension culture,the lethal concentrations of geneticin were 60 mg/L and 30 mg/L,respectively.【Conclusion】The tolerance concentrations of geneticin for Chinese chestnut embryogenic callus under solid and suspension culture were determined.This study lays a foundation for the use of geneticin in the chestnut genetic transformation.
