钝叶酢浆草的离体培养与快速繁育研究 被引量:4

摘要 【目的】构建钝叶酢浆草组培快繁的技术体系。【方法】以3种钝叶酢浆草品种的叶片和茎尖为外植体,设置不同质量浓度的激素组合,筛选最适合钝叶酢浆草培养的启动培养基、继代培养基和生根培养基。【结果】钝叶酢浆草最适外植体为叶片,最适消毒方法为用75%酒精和2%NaClO溶液浸泡10 min,最适启动培养基为MS+6-BA 1.5 mg/L+NAA 0.2 mg/L,28 d后出愈率达51.8%,最适继代培养基为MS+6-BA 2.0 mg/L+NAA 0.1 mg/L,最适生根培养基为1/2 MS+NAA 0.1 mg/L。3种钝叶酢浆草中最适合组织培养操作的品种是钝叶酢浆草‘大花’(Oxalis obtusa‘large form’)。【结论】确立钝叶酢浆草组织培养技术体系,获得了无菌苗,为批量化生产提供基础。 【Objective】Oxalis obtusa belong to herbaceous plant.To rapidly produce excellent varieties of Oxalis obtusa,explant leaves were used to establish tissue culture system.【Methods】To screen the most suitable starting culture medium,subculture medium and rooting medium,we compared the growing status of plants in different concentrations of hormones.【Results】In these three cultivars,the best cultivar in technical culture operation was Oxalis obtuse‘large form’.The best explant was leaf.The most appropriate disinfection method was 75%alcohol+2%NaClO liquor soaked 10 minutes.MS medium supplemented with 6-BA 1.5 mg/L+NAA 0.2 mg/L was the best starting culture medium with 51.8%induction rate in 28 days.The most suitable subculture medium and rooting medium were MS+6-BA 2.0 mg/L+NAA 0.1 mg/L and 1/2 MS+NAA 0.1 mg/L,respectively.【Conclusion】In this study,we established technical culture system for Oxalis obtuse and obtained tissue culture seedlings.The results will provide the basis for industrial production.
