布鲁氏菌Omp2b蛋白抗原表位的生物信息学分析 被引量:3

摘要 目的利用生物信息学软件分析布鲁氏菌外膜蛋白2b(Omp2b)蛋白的抗原表位。方法从NCBI数据库中获取羊布鲁氏菌Omp2b蛋白的氨基酸序列。通过ProtParam软件分析Omp2b蛋白的理化性质;利用SOMPA在线软件分析Omp2b蛋白的二级结构;使用SWISS-MODEL在线分析和构建Omp2b蛋白的三级结构,并使用RasMol2.7.2.1软件分析。利用IEDB、DNAStar软件预测Omp2b蛋白的B细胞抗原表位;利用SYFPEITHI、IEDB软件预测Omp2b蛋白的T细胞抗原表位。结果对Omp2b蛋白的一级结构分析发现,Omp2b蛋白是一种稳定亲水性蛋白质;在Omp2b蛋白的二级结构中,无规卷曲和β转角构成较多;Omp2b蛋白的三级结构是环状的,非常稳定,其表位主要位于三级结构的上部和下部。经DNAStar、IEDB软件预测,Omp2b蛋白具有8个B细胞抗原表位,其位点分别位于66~85、115~131、169~176、184~188、196~212、268~286、306~316、328~352。经SYFPEITHI、IEDB在线软件预测,Omp2b蛋白具有7个CTL优势表位,其位点分别位于76~84、102~110、231~239、241~249、290~298、329~337、335~343;Omp2b蛋白具有3个Th优势表位,其位点分别位于133~147、178~192、202~216。结论布鲁氏菌Omp2b蛋白具有7个CTL优势表位、3个Th优势表位和8个B细胞抗原表位,具有良好的免疫原性。这为布鲁氏菌病诊断和疫苗研发提供了一定理论依据。 Objective Using bioinformatics software to analyze the epitope of Brucella outer membrane protein 2b(Omp2b)protein.Methods The amino acid sequence of Brucellosis Omp2b was obtained from NCBI database.Physicochemical properties of Omp2b protein were analyzed by ProtParam software.The secondary structure of Omp2b protein was analyzed by SOMPA.Swiss-model was used to analyze and construct the tertiary structure of Omp2b protein online,and RasMol software was used for analysis.IEDB and DNAStar software were used to predict the B-cell epitopes of Omp2b protein.SYFPEITHI and IEDB software were used to predict the T-cell epitopes of Omp2b protein.Results The primary structure analysis of Omp2b protein found that Omp2b protein was a stable hydrophilic protein.In the secondary structure of Omp2b,there were more random crimp andβ-turn.The tertiary structure of Omp2b protein was barrel-shaped and stable,and its epitopes were mainly located at the upper and lower end of the tertiary structure.According to the prediction by DNAStar and IEDB software,Omp2b protein had 8 B cell antigen epitopes,and its loci were located in 66-85,115-131,169-176,184-188,196-212,268-316,and 328-352.According to the prediction of SYFPEITHI and IEDB,Omp2b protein had 7 CTL dominant epitopes,and the loci were located in 76-84,102-110,231-239,241-249,290-298,329-337,and 335-343,respectively;Omp2b protein had 3 Th dominant epitopes,and the sites were located in 133-147,178-192,and 202-216,respectively.Conclusion Omp2b protein has 7 CTL dominant epitopes,3 Th dominant epitopes and 8 B cell epitopes,indicating that Omp2b protein has good immunogenicity,which provides a theoretical basis for the diagnosis of brucellosis and vaccine development.
