摘要
目的建立能同时测定消渴灵片中毛蕊花糖苷、马替诺皂苷、五味子醇甲、麦冬甲基黄烷酮A、五味子甲素、甲基麦冬二氢高异黄酮B和五味子乙素含量的HPLC法。方法采用Kromasil C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相:乙腈-1 mL·L^(-1)冰醋酸,梯度洗脱;流速:1.0 mL·min^(-1);柱温:30℃;检测波长分别为330(检测毛蕊花糖苷和马替诺皂苷),296(检测麦冬甲基黄烷酮A和甲基麦冬二氢高异黄酮B)和217 nm(检测五味子醇甲、五味子甲素和五味子乙素)。结果 7种成分分别在4.67~116.75,2.39~59.75,1.46~36.50,0.93~23.25,1.54~38.50,0.58~14.50和1.26~31.50μg·mL^(-1)范围内线性关系良好(r≥0.999 2);平均回收率分别为99.80%,98.11%,97.92%,98.23%,100.01%,96.99%和97.89%;RSD值分别为0.96%,1.33%,1.40%,0.71%,0.82%,1.11%和1.05%。结论该方法操作简便、重复性好,可作为消渴灵片的质量控制方法。
Objective To establish an HPLC method for the simultaneous determination of verbascoside,martinoside,schisandrin,methylophiopogonanone A,deoxyschizandrin,methylophiopogonanone B andγ-schisandrin in Xiaokeling Tablets.Methods The chromatographic separation was carried out on a Kromasil C 18 column(250 mm×4.6 mm,5μm)with the mobile phase consisting of acetonitrile-1 mL·L-1 acetic acid with gradient elution at a flow rate of 1.0 mL·min-1.The column temperature was 30℃.The detection wavelength of verbascoside and martinoside was 330 nm,the detection wavelength of methylophiopogonanone A and methylophiopogonanone B was 296 nm,and the detection wavelength of schisandrin,deoxyschizandrin andγ-schisandrin was 217 nm.Results The 7 components showed good linear relationships within the ranges of 4.67-116.75,2.39-59.75,1.46-36.50,0.93-23.25,1.54-38.50,0.58-14.50 and 1.26-31.50μg·mL-1(r≥0.9992),whose average recoveries were 99.80%,98.11%,97.92%,98.23%,100.01%,96.99%and 97.89%with the RSDs of 0.96%,1.33%,1.40%,0.71%,0.82%,1.11%and 1.05%,respectively.Conclusion The method is easy to operate and repeatable.It can be valued as a quality control method for Xiaokeling Tablets.
