摘要
开展西藏自治区林周县亏组山草地土壤丝状真菌多样性研究,探究围栏封育和海拔对土壤理化因子及土壤丝状真菌多样性的影响。采用稀释涂布平板法从亏组山13个样点中分离丝状真菌,应用ITS rDNA序列分析法对获得的菌株进行分类鉴定,运用R 4.0.1和SPSS 20.0等软件对丝状真菌多样性及其与环境因子相关性进行分析。结果显示,从亏组山草地土壤中分离得到1244株丝状真菌,分属于23个属37个种,其中围栏内733株,分属于21个属35个种,围栏外511株,分属于23个属35个种。出现频率和相对丰度显示,各样点优势种为Pseudogymnoascus roseus和Mucor hiemalis,围栏内优势种为Pseudogymnoascus roseus和Mucor hiemalis,围栏外的优势种为Pseudogymnoascus roseus和Penicillium sp.。统计结果表明,围栏封育对土壤理化因子以及土壤丝状真菌多样性指数的影响不明显,放牧干扰一定程度上影响了土壤丝状真菌对土壤理化因子的响应程度。
The filamentous fungi diversity in Kuizu mountain,Linzhou County and the influence of environmental factors on bacteria community were analyzedby using R 4.0.1 SPSS 20.0 Filamentous fungi were isolated by dilution plate method and the obtained strains were classified and identified with ITS rDNA sequence analysis.The result showed that total 1244 filamentous fungi isolated were identified to 23 genera and 37 species.Pseudogymnoascus roseus and Mucor hiemalis were the dominate species in the Kuizu mountain.Total of 733 filamentous fungi isolateed were identified to 21 genera and 35 species from inside the fence,among them Pseudogymnoascus roseus and Mucor hiemalis were the dominate species,and total of 511 filamentous fungi isolated were identified to 23 genera and 35 species from outside the fence,among them Pseudogymnoascus roseus and Penicillium sp.were the dominated species.In general,long-term grazing exclosure had no significant effects on soil nutrients and soil filamentous fungi community diversity in the study area.Fencing affected the relationship between grassland soil filamentous fungi and soil environmental factors at different altitudes.
