九香虫血淋巴不同保存温度下的稳定性评价 被引量:3

摘要 九香虫是一种传统药用昆虫,其血淋巴具有抗癌、抗菌作用,也可用以治疗婴儿血管瘤等,其抗癌活性物质主要为多肽。但九香虫血淋巴在不同保存条件下稳定性较差,易产生不溶性沉淀,导致其总蛋白含量变化以及抗癌活性变动。本文通过对常规保存温度(4、-20、-80℃)下的九香虫血淋巴产生不溶性沉淀及总蛋白含量的测定,并通过体外MTT法,检测不同处理的九香虫血淋巴的体外抗癌活性,以评价九香虫血淋巴的稳定性。结果显示,-80℃下保存的血淋巴产生不溶性沉淀量相对较少,总蛋白含量在14 d首次取出无显著变化,不同处理的九香虫血淋巴均能明显抑制乳腺癌细胞的生长,-80℃抗癌活性相对稳定。综上,-80℃温度下较适合九香虫血淋巴的保存。 Aspongopus chinensis is a traditional medicinal insect,and its hemolymph has anti-cancer and anti-bacterial effects.It can also be used to treat hemangioma etc.But A.chinensis hemolymph is less stable under different storage conditions,and it is easy to produce insoluble precipitation,which leads to changes in its total protein content and anti-cancer activity.In this paper,the total protein content and precipitation of the hemolymph under different temperatures(4,-20,-80℃)were observed continuously.The anti-cancer activity in vitro of the hemolymph with different treatments by MTT method,and the hemolymph stability of A.chinensis was evaluated.The results showed that the amount of insoluble precipitation produced by hemolymph was relatively small when stored at-80℃.Total protein content was not significantly changed when the hemolymph was first removed after 14 days.The hemolymph treated with different treatments could significantly inhibit the growth of breast cancer cells,and the anticancer activity was relatively stable at-80℃.To sum up,the temperature of-80℃is more suitable for the preservation of hemolymph of A.chinensis.
