如意伤科膏的配方筛选及体外评价

摘要 目的:筛选如意伤科膏的配方组成,测定指标成分含量,并评价其体外经皮渗透特性。方法:采用正交试验设计,以物理性状为指标,参考君药栀子的主要有效成分栀子苷含量筛选如意伤科膏配方,选用Waters Symmetry RP18色谱柱(250mm×4.6mm,5μm),以乙腈-水(25∶75)为流动相,检测波长为238nm,采用Franz扩散池分别开展如意伤科膏及药粉传统外敷用法的体外经皮渗透试验。结果:如意伤科膏的最佳配方为凡士林∶羊毛脂∶液体石蜡∶(20∶1∶2),栀子苷的含量为0.2372mg/g,24h累计透皮量为0.0389mg/g,累计透皮率为16.41%。结论:筛选的配方能满足使用需求,如意伤科膏的体外透皮性能明显优于传统外敷形式。 Objective:To screen the formulation of Ruyi Injury Ointment,determine the content of index components,and evaluate its percutaneous permeability in vitro.Methods:The formulation of Ruyi Injury Ointment was screened by orthogonal design,taking physical properties as index and referring to the content of gardenoside,which was the main active ingredient of Gardenia jasminoides.Waters Symmetry RP18 column(250 mm×4.6 mm,5 m)was selected,acetonitrile-water(25:75)was used as mobile phase,and the detection wavelength was 238 nm.The traditional external application of Ruyi Injury Ointment and Powder was carried out by Franz diffusion cell and external percutaneous absorption test.Results:The optimum formula of Ruyi Injury Ointment was Vaseline:Lanolin:Liquid Paraffin:20:1:2,the content of Geniposide was 0.2372 mg/g.The cumulative transdermal volume was 0.0389 mg/g in 24 hours,and the cumulative transdermal rate was 16.41%.Conclusion:The selected formulation can meet the needs of application.The in vitro transdermal performance of Ruyi Injury Ointment is obviously superior to that of traditional external application.
