摘要
目的研究推拿点按手法对慢性压迫性神经损伤模型(CCI)大鼠镇痛作用及对其背根神经节(DRG)PKC及P2X3表达的影响。方法将80只SD大鼠随机分为4组(每组20只):空白组、模型组、假手术组、造模+点按组。采集各组大鼠术前及术后第1,3,7,14,21天触诱发痛的测定(von Frey测试)指标,并检测点按手法干预结束后各组大鼠DRG神经元PKC及P2X3的表达情况。结果(1)造模后,与空白组和假手术组比较,模型组和造模+点按组大鼠的触诱发痛在术后即开始下降(P<0.01)。与模型组比较,造模+点按组大鼠的触诱发痛在术后1~7 d的差别无统计学意义(P>0.05),至术后14 d开始上升(P<0.05),至术后21 d差别更为明显(P<0.01)。(2)与空白组和假手术组比较,模型组大鼠DRG中PKC和P2X3的表达水平显著增高(P<0.01);手法干预后,造模+点按组PKC表达水平低于模型组(P<0.05)、高于空白组和假手术组(P<0.05);P2X3受体的表达水平低于模型组(P<0.01)、高于空白组和假手术组(P<0.05)。结论与空白组、假手术组比较,坐骨神经CCI模型大鼠的DRG细胞存在PKC和P2X3过表达,PKC及P2X3与神经病理性疼痛相关,推拿手法干预可抑制CCI模型大鼠DRG中PKC及P2X3的表达,发挥镇痛作用。
Objective To study the analgesic effect of Dianan massage on chronic constriction injury(CCI)rats and its effect on the expression of PKC and P2X3 in dorsal root ganglia.Methods 80 SD rats were randomly divided into 4 groups(20 rats in each group),which was blank group,model group,sham operation group,model+Dianan group,respectively.The indexes of pain induced by touch(von Frey test)were collected before operation and at day 1,3,7,14,21 after operation,and the expression of PKC and P2X3 in DRG neurons of rats in each group was detected after the intervention.Results Compared with the blank group and the sham operation group,the induced pain in the model group and the model+Dianan group began to decrease after operation(P<0.01);compared with the model group,the induced pain in the model+Dianan group had no significant difference between 1-7 days after operation(P>0.05),and increased from 14 days after operation(P<0.05),and reached a significant difference at day 21(P<0.01).Compared with the blank group and the sham operated group,the expression level of PKC and P2X3 in DRG in the model group was significantly higher(P<0.01).After manual intervention,the expression level of PKC in the model+Dianan group was lower than that in the model group(P<0.05),higher than that in the blank group and the sham operated group(P<0.05),and the expression level of P2X3 receptor was lower than that in the model group(P<0.01),higher than that in the blank group and the sham operated group(P<0.05).Conclusion Compared with the blank group and the sham operated group,there were over expression of PKC and P2X3 in DRG cells of sciatic nerve CCI model rats,which proves that PKC and P2X3 are related to neuropathic pain.Massage intervention can inhibit PKC and P2X3 expression in DRG of CCI model rats and play an analgesic role.
