摘要
目的建立用于快速筛查未知药膏中是否含有利多卡因、盐酸普鲁卡因、盐酸丁卡因3种常用局部麻醉药品的HPLC-DAD方法。方法采用HPLC-DAD法,色谱柱:C 18柱(4.6×150mm,5μm);柱温:30℃;流动相:甲醇-水(含0.4%三乙胺,用磷酸调节pH值至6.5)(60∶40)。结果比较3种目标分析物与已知对照品的保留时间及三维光谱图,对未知药膏中的麻醉药品进行定性,检测出大多数未知药膏中含有利多卡因与盐酸丁卡因。结论采用HPLC-DAD法检验未知药膏中是否含有利多卡因、盐酸普鲁卡因、盐酸丁卡因,为公安局及市场监督管理局查处市场上非法麻醉药膏提供技术支持。
OBJECTIVE To establish an efficient method by HPLC-DAD for identifying lidocaine,procaine hydrochloride and tetracaine hydrochloride which are commonly used anesthetics in unknown cream.METHODS Analyzing by HPLC-DAD,stationary phase:C 18 column(4.6×150mm,5μm);column temperature:30℃;Mobile phase:methanol-water(containing 0.4%triethyla mine,)pH adjusted to 8.0 with phosphoric acid(60∶40).RESULTS Lidocaine and tetracaine hydrochloride were detected in most of the unknown cream by comparing of retention time and 3D spectrum with the three target analytes and reference substance and characterizing anesthetics.CONCLUSION The HPLC-DAD method can be used to test whether the unknown cream contains lidocaine,procaine hydrochloride and tetracaine hydrochloride,and provide technical support for the Public Security Bureau and the Market Supervision Ad ministration to investigate illegal anesthetic ointments on the market.
