三七总皂苷和多烯磷脂酰胆碱调控大鼠非酒精性脂肪肝的药效学比较 被引量:2

Pharmacodynamic comparison of panax notoginseng saponins and polyene phosphatidylcholine in regulating nonalcoholic fatty liver in rats

摘要 目的对比三七总皂苷和多烯磷脂酰胆碱对大鼠非酒精性脂肪肝(NAFLD)的药效作用,进一步探讨三七总皂苷治疗NAFLD的作用机制。方法将雄性SD大鼠随机分为空白对照组(Blank,n=8)及高脂饮食组(HFD,n=24)。Blank组以普通饲料饲养,HFD组以高脂饲料进行饲养,共饲养4周。将HFD组大鼠随机分为三组,依次为模型组(Model,n=8)、三七总皂苷组(PNS,n=8)、多烯磷脂酰胆碱组(PPC,n=8)。分组后按要求给予药物或生理盐水灌胃4周。PNS组给药剂量为0.2g/(kg·d),PPC组折算后给药浓度为0.1g/(kg·d),Blank组及Model组均给予对应体积的生理盐水。检测血清天门冬氨酸氨基转移酶(AST),丙氨酸氨基转移酶(ALT)、碱性磷酸酶(ALP)、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)等的变化。结果三七总皂苷组的体重值、肝重指数、肝功能指标(AST、ALT和ALP值)、血脂指标(LDL-C、TC、TG值)都较低,与模型组相比差异有统计学意义(P<0.05)。多烯磷脂酰胆碱组仅体重、肝功能指标仅ALT值、血脂指标仅TG值与模型组比较,差异有统计学意义(P<0.05)。结论三七总皂苷在NAFLD大鼠的治疗中具有降低体重,降低血脂,改善肝功能指标等作用,与目前临床广泛用于护肝的药物多烯磷脂酰胆碱相比,三七总皂苷具有综合优势。 Objective To compare the pharmacodynamic efficacies of panax notoginseng saponins and polyene phosphatidylcholine in regulating nonalcoholic fatty liver disease(NAFLD)in rats,and to further investigate the mechanism of panax notoginseng saponins on NAFLD.Methods Male SD rats were randomly divided into the blank group(Blank,n=8)and the high fat diet group(HFD,n=24).The blank group was fed with normal diet,while the HFD group was fed with high fat diet for 4 weeks.Rats in the HFD group were randomly divided into three groups,which were the model group(Model,n=8),the panax notoginseng saponins group(PNS,n=8)and the polyene phosphatidylcholine group(PPC,n=8).After grouping,they were given drugs or normal saline for 4 weeks.The dose of the PNS group was 0.2g/(kg·d),and the converted concentration of the PPC group was 0.1g/(kg·d).Both the blank group and the model group were given corresponding volumes of normal saline.Changes of serum aspartate aminotransferase(AST),alanine aminotransferase(ALT),alkaline phosphatase(ALP),total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C)were detected.Results Body weight,liver weight index,liver functions of AST value,ALT value and ALP value,LDL-C value,TC value and TG value of blood lipid in the panax notoginseng saponins group were lower than those in the model group(P<0.05).There were significant differences between the polyene phosphatidylcholine group and the model group in weight,liver function index of ALT value and TG value of blood lipid index(P<0.05).Conclusion The panax notoginseng saponins reduces the body weight,decreases the blood lipid and improves the liver function indexes in NAFLD rats.Compared with polyene phosphatidylcholine,which is widely used for liver protection in clinic,panax notoginseng saponins have comprehensive advantages.
