蜕膜中核心聚糖蛋白的表达水平与发生稽留流产的关系

The relationship between the expression of decorin and missed abortion in decidua

目的探讨蜕膜组织中核心聚糖蛋白(DCN)与稽留流产发生的关系。方法选取广州医科大学附属第三医院2019年1-9月孕5~10周的19例稽留流产患者(稽留流产组)的蜕膜组织和18例行选择性早期人工流产患者(对照组)的蜕膜组织,采用单细胞转录组测序分析蜕膜基质细胞中DCN表达;采用实时荧光定量PCR法比较蜕膜组织中DCN m RNA的表达水平;免疫组化法检测DCN在蜕膜组织的定位;免疫印迹法比较DCN蛋白表达水平。结果两组孕妇年龄、妊娠周数及孕次比较差异无统计学意义(P>0.05);单细胞转录组测序发现,DCN在蜕膜基质细胞中高表达,与对照组比较,稽留流产组蜕膜基质细胞DCN表达增加,差异有统计学意义(P<0.0001);实时荧光定量PCR及免疫印迹法结果显示稽留流产组DCN mRNA表达水平及蛋白表达水平均明显升高(P<0.01);免疫组化结果显示两组蜕膜细胞形态无明显差异,DCN蛋白在蜕膜组织中广泛存在,蜕膜基质细胞的胞质及胞间均有表达。结论稽留流产组蜕膜组织中DCN高表达,可能与稽留流产发生相关。

Objective To investigate the relationship between the expression of decorin(DCN)in decidua and the occurrence of missed abortion.Methods The decidua tissues of 19 cases of missed abortion(missed abortion group)and 18 cases of selective early abortion(control group)were collected in the Third Affiliated Hospital of Guangzhou Medical University from January to September 2019.DCN mRNA expression in different cell type of decidua was deteced by single cell transcript sequencing;the expression level of DCN mRNA in decidua tissue were detected by real-time fluorescence quantitative PCR;the localization of DCN in decidua were detected by immunohistochemical staining;the expression level of DCN protein were detected by Western blot.Results There was no significant difference in age,gestational weeks and gestational times between two groups(P>0.05).The result of single cell transcriptome sequencing showed that DCN was highly expressed in decidual stromal cells.Compared with the control group,the expression of DCN in decidual stromal cells of the missed abortion group increased significantly(P<0.0001);the results of RT-qPCR and Western blotting showed that the expression of DCN at mRNA and protein level in the group of missed abortion was significantly increased(P<0.01);the results of immunohistochemistry showed that there was no significant difference in the morphology of decidual cells between the two groups.DCN protein was widespread in decidual tissues and located both in cytoplasm and intercellular of decidual stromal cells.Conclusion The high expression of DCN in decidua might be related to the occurrence of missed abortion.
