摘要
以红曲米来源的5株红曲菌(M1~M5)为研究对象,对其进行形态观察、生理生化试验及分子生物学鉴定,并采用超高液相色谱串联质谱(UPLC-MS/MS)法测定洛伐他汀及桔霉素产量,筛选高产洛伐他汀低产桔霉素的优良菌株。在此基础上,利用该菌株研制功能红腐乳,并结合感官指标、理化指标及微生物指标等对其品质进行评价。结果表明,5株红曲菌中菌株M1、M3、M4、M5均被鉴定为紫色红曲霉(Monascus purpureus),菌株M2被鉴定为丛毛红曲霉(Monascus pilosus)。其中,菌株M2发酵液中洛伐他汀产量最高(15.67 mg/L),确定其为高产洛伐他汀的优良菌株。利用菌株M2研制的功能红腐乳的质量指标均满足相关国标要求。
Five strains of Monascus sp.(M1-M5)from red kojic rice were identified by morphological observation,physiological and biochemical tests and molecular biology techniques,and the production of lovastatin and citrinin was determined by UPLC-MS/MS,in order to screen excellent strain with high-producing lovastatin and low-producing citrinin.On this basis,the functional red sufu was developed using the strain,and its quality was evaluated by sensory,physical and chemical,and microbial indexes.The results showed that the strains M1,M3,M4,and M5 were all identified as Monascus purpureus,and the strain M2 was identified as Monascus pilosus.Among them,the production of lovastatin in the fermentation broth of strain M2(15.67 mg/L)was the highest,which was an excellent strain with high-yield lovastatin.The quality indexes of the functional red sufu developed by strain M2 met the requirements of relevant national standards.
