白首乌酒对癫痫大鼠神经元保护作用及其机制研究 被引量:3

摘要 探讨白首乌酒对癫痫大鼠神经元保护作用及其机制。50只SD大鼠随机分为空白组、模型组、白首乌酒提取物(CBLE)低剂量组(4 mL/kg)、高剂量组(10 mL/kg)。除空白组外其他组大鼠进行癫痫模型的建立,建模成功后进行Racine评分、记录癫痫发作次数及首次癫痫发作持续时间;通过苏木精-伊红(HE)染色法、切片扫描,观察大鼠神经元损伤情况;蛋白免疫印迹(Western bolt)分析大鼠大脑海马组织中p38MAPK蛋白表达量。结果表明,CBLE各剂量组Racine评分、癫痫发作次数和首次癫痫发作持续时间与模型组相比显著降低(P<0.05)、海马神经细胞紧密性有明显提高;CBLE各剂量组p38MAPK/GADPH比值与模型组相比都有差异性(P<0.05)。结果表明,高剂量的CBLE可以减轻癫痫症状,对癫痫大鼠神经元具有保护作用,这种保护作用是通过抑制p38MAPK蛋白的表达实现的。 To investigate the protective effect and mechanism of Cynanchum bungei liquor on neurons in epileptic rats,50 SD rats were randomly divided into blank group,model group,C.bungei liquor extract(CBLE)low-dose group(4 ml/kg)and high-dose group(10 ml/kg).Except for the blank group,the epileptic model was established in the other groups,and Racine score,epilepsia seizure frequency and first epilepsia seizure duration were performed after successful modeling.The damage of rat neurons was observed by hematoxylin-eosin(HE)staining and section scanning.The expression of p38MAPK protein in hippocampus of rat brain was analyzed by Western Bolt.The results shown that it was found that compared with the model group,the Racine score,epilepsia seizure frequency and first epilepsia seizure duration of each dose groups of CBLE were reduced(P<0.05),and the compactibility of hippocampal nerve cells of rats was significantly improved.The p38MAPK/GADPH ratio of each dose groups of CBLE was significantly different from that of the model group(P<0.05).The results showed that high dose of CBLE not only alleviated epilepsy symptoms,but also had a protective effect on epileptic rat neurons,which was achieved by inhibiting the expression of p38MAPK protein.
