泻肺利水颗粒对自发性高血压大鼠肾素-血管紧张素-醛固酮系统的影响 被引量:4

摘要 目的研究泻肺利水颗粒对自发性高血压大鼠血清血管紧张素Ⅰ、血管紧张素Ⅱ、血管紧张素转换酶、肾素、醛固酮水平的影响,探讨泻肺利水颗粒的降压机制。方法将自发性高血压大鼠50只(13周龄)随机分为模型组、中药高剂量组、中药中剂量组、中药低剂量组、卡托普利组,每组10只;空白对照组为10只Wistar-Kyoto大鼠(WKY大鼠)。分别测定治疗前和4周后的大鼠尾动脉压,以及4周后血清血管紧张素Ⅰ、血管紧张素Ⅱ、血管紧张素转换酶、肾素、醛固酮水平,进行统计分析。结果与模型组比较,中药高、中、低剂量组及卡托普利组4周后血压明显降低(P<0.01);4周后血压,中药中剂量组与卡托普利组相比,差异无统计学意义(P>0.05)。与模型组相比,中药高、中、低剂量组及卡托普利组血清肾素、血管紧张素Ⅰ水平皆明显降低,差异有统计学意义(P<0.01)。与模型组相比,中药中剂量组、卡托普利组血清醛固酮、血管紧张素Ⅱ水平明显偏低,差异有统计学意义(P<0.01),中药高剂量、低剂量组醛固酮、血管紧张素Ⅱ水平偏低,差异有统计学意义(P<0.05)。各组间血管紧张素转换酶水平比较差异无统计学意义(P>0.05)。结论泻肺利水颗粒能够显著降低自发性高血压大鼠尾动脉压,其降压机制可能与抑制肾素-血管紧张素-醛固酮系统相关。 Objective To investigate the effect of Xiefei Lishui Granules on spontaneous hypertension rats(SHR) through testing the level of Ang I,Ang II,ACE,renin and aldosterone(ALD)by ELISA to explore the mechanism of anti-hypertensive effects of Xiefei Lishui Granules.Methods 50 SHR of 13-week old were randomly divided into model group,high dose,medium dose,and low dose of Chinese medicine group and captopril group,with 10 rats in each group;10 Wistar-Kyoto(WKY) rats as normal control group.The blood pressure of the tail artery of rats was measured before the experiment and 4 weeks later.The level of Ang I,Ang II,ACE,renin and ALD were tested and analyzed respectively by ELISA after 4 weeks.Results Compared with the model group,the blood pressure of high dose group,medium dose group,low dose group of Chinese herbal medicine and captopril group were decreased after 4 weeks(PP<0.01).The blood pressure of medium dose group and captopril group showed no significant statistical difference after 4 weeks.Compared with the model group,the level of serum renin and Ang I of captopril group,high dose group,medium dose group and low dose group of Chinese herbal medicine were significantly decreased with the difference statistically significant(P<0.01).Compared with the model group,the level of ALD and Ang II of captopril group and medium dose group were much lowered with the difference statistically significant(P<0.01),the level of ALD and Ang II of the high dose group and low dose group of Chinese medicine were lowered with the difference statistically significant(P<0.05).The level of Angiotensin converting enzyme(ACE) showed no difference between the groups.Conclusion Xiefei Lishui Granules can effectively reduce the blood pressure of tail artery of SHR,and the mechanism might be the effect of inhibiting of the renin-angiotensin-aldosterone system.
