逍遥丸对产前应激子代大鼠学习认知的影响机制

摘要 目的:探讨逍遥丸对产前应激(prenatal stress,PS)子代大鼠学习认知的影响及其可能作用机制。方法:本实验研究时间为2018年9月-2019年11月,选择40只SD大鼠,建立PS子代大鼠模型,根据是否应激、灌胃不同药物,将大鼠分为CON组、PS组、PS+Xiaoyao Pill组和PS+NS组,各10只。通过Morris水迷宫观察各组大鼠的学习认知行为,再运用Western blot观察各组大鼠海马体中GluA1蛋白表达的变化。结果:PS组到达平台所用时间明显长于CON组,而PS+Xiaoyao Pill组到达平台时间明显短于PS+NS组,差异均有统计学意义(P<0.05)。PS组GluA1蛋白的相对表达量低于CON组,而PS+Xiaoyao Pill组的GluA1蛋白相对表达量高于PS+NS组,差异均有统计学意义(P<0.05)。结论:逍遥丸可有效改善PS子代大鼠的学习认知功能,与其发挥作用机制之一可能是提高大鼠海马体GluA1蛋白的表达有关。 Objective:To investigate the effect of Xiaoyao Pills on learning and cognitive of rats under prenatal stress,and its possible mechanism of action.Method:The research time of this experiment is September 2018 to November 2019,40 SD rats were selected and PS offspring rat model was established,according to different stress and gavage drugs,the rats were divided into CON group,PS group,PS+Xiaoyao Pill group and PS+NS group,10 rats in each group.The learning and cognitive behaviors of rats in Morris water maze were observed,the changes of GluA1 protein expression in the hippocampus of rats in each group were observed by Western blot.Result:The time to reach the platform in PS group was significantly longer than that in CON group,while the time to reach the platform in PS+Xiaoyao Pill group was significantly shorter than that in PS+NS group,with statistically significant differences(P<0.05).GluA1 relative expression in PS group was lower than that in CON group,while GluA1 relative expression in PS+Xiaoyao Pill group was higher than that in PS+NS group,with statistically significant differences(P<0.05).Conclusion:Xiaoyao Pills can effectively improve the learning and cognitive function of PS offspring rats,one of its mechanisms may be related to the enhancement of GluA1 protein expression in rat hippocampus.
