摘要
目的:研究甲状腺乳头状癌(PTC)组织自噬相关蛋白LC3与Beclin1及其调控基因的表达与分析。方法:选择20例手术切除的PTC新鲜组织,另外收集同期因甲状腺良性病变切除的正常甲状腺新鲜组织标本20例。分别采用Western Blot法检测PTC组织(恶性)和正常甲状腺组织(良性)中LC3与Beclin1蛋白的表达情况,采用实时荧光定量聚合酶链反应(RT-PCR)测定PTC组织和正常甲状腺组织中LC3A、LC3B以及Beclin1蛋白基因(BECN1)mRNA相对表达量。结果:RT-PCR结果显示,PTC组织中的LC3B mRNA相对表达高于良性病变甲状腺组织(P<0.05);Western Blot结果显示自噬相关蛋白Beclin1表达差异无统计学意义(P>0.05),但是LC3蛋白的亚型蛋白LC3B在恶性肿瘤组织中表达显著上调(P<0.05)。结论:LC3B蛋白可能是PTC的靶蛋白,在甲状腺肿瘤良恶性的诊断过程中应得到重视。
Objective:To study the expression and analysis of autophagy related proteins LC3,Beclin1 and their regulatory genes in papillary thyroid carcinoma.Method:A total of 20 fresh resected PTC specimens were collected from the hospitalized patients,and other contemporaneous 20 normal thyroid tissue specimens were collected from the resected thyroid benign lesions.The expression levels of LC3 and Beclin1 protein in PTC tissues and normal thyroid tissues were detected by Western Blot.The real-time fluorescence quantitative polymerase chain reaction(RT-PCR)was used to determine the relative expression levels of LC3A、LC3B and BECN1 mRNA in PTC tissues and normal thyroid tissues.The expression level of LC3 and Beclin1 protein,LC3A,LC3B and BECN1 mRNA and between PTC and normal thyroid tissues was investigated.Result:The RT-PCR results showed that the LC3B mRNA expression in the PTC tissue was significantly higher than that in the normal thyroid tissue(P<0.05).The Western Blot results showed that the expression of Beclin1 protein in the PTC tissue was higher than that in the normal thyroid tissue with no significant difference(P>0.05),however,the expression of LC3B was significantly higher than that in the normal thyroid tissue(P<0.05).Conclusion:LC3B protein may be the target protein in PTC,more attention should be paid to the diagnosis of benign and malignant thyroid tumors.
