摘要
论文主要运用紫外可见分光光度法,以裙带菜中有效成分褐藻胶含量为指标,对提取温度、提取时间和料液比三个不同因素进行考察,确定裙带菜多糖的最佳提取工艺.根据极差分析R值可知,各因素影响多糖得率效果的主次顺序为提取温度>提取时间>料液比,通过方差分析验证提取温度为显著性因素.故确定裙带菜多糖的最佳提取工艺条件为提取时间8 h,提取温度70℃,料液比1∶60.通过方法学考察可知,本实验所采用的裙带菜多糖含量测定方法稳定可靠,具有较好的精确度、稳定性、重现性和准确性,可用于裙带菜多糖提取的含量测定.
In this thesis,the use of ultraviolet and visible spectrophotometry to the active ingredient in seaweed alginate content index,extraction temperature,extraction time and solid to liquid ratio of three different factors were investigated to determine the best seaweed polysaccharide extraction.Known under the range of Rvalues,the factors affecting the yield of polysaccharide primary and secondary order effect of extraction temperature>extraction time>liquid ratio,extraction temperature verified by analysis of vari⁃ance as a significant factor.Therefore,to determine the best seaweed polysaccharide extraction conditions for the extraction time 8 h,extraction temperature 70℃,liquid ratio 1∶60.Through the research method⁃ology,it is known that the determination method of seaweed polysaccharides used in this study is stable and reliable,has good accuracy,stability,reproducibility and accuracy,can be used for the wakame deter⁃mination of polysaccharides.
