鱼源泡囊短波单胞菌16S rDNA基因及耐药性分析 被引量:1

Analysis of 16S rDNA Gene and Drug Resistance of Brevundimonas vesicularis in Fish

摘要 为了鉴定从黄颡鱼中分离的泡囊短波单胞菌并探究其耐药性,用PCR方法和生物信息学方法分析16S rDNA基因,用PCR方法检测该菌株的3种磺胺类、4种氨基糖苷类、1种β-内酰胺类耐药基因,用纸片琼脂扩散法进行药敏试验。结果表明,菌株16S rDNA序列长度为1372 bp,GenBank登录号为MK294348,与泡囊短波单胞菌一致性最高(98%),系统进化树显示与泡囊短波单胞菌266XY4株聚为一类,从而判定该菌为泡囊短波单胞菌。该菌株具有磺胺类耐药基因Sul1、氨基糖苷类耐药基因aac(6′)-Ⅰb、β-内酰胺类耐药基因TEM。该菌对哌拉西林、卡那霉素、氨苄西林等抗生素均有耐药性,药敏试验结果与耐药基因检测结果基本一致,为研究泡囊短波单胞菌的耐药机制提供了参考资料。 In order to identify Brevundimonas vesiculari strain isolated from diseased Pelteobagrus fulvidraco and explore its drug resistance,the Brevundimonas vesiculari strain was identified by 16S rDNA using PCR.Drug resistance genes of three sulfonamides,four aminoglycosides and oneβ-lactam were detected by PCR.Drug sensitivity test was detectd by Kirby-Bane for isolated strains.The identification results showed that the length of 16S rDNA sequence was 1372 bp(GenBank accession No.MK294348),which was the highest consistency with Brevundimonas vesicularis(98%).Phylogenetic tree showed that the strain FBV was clustered with Brevundimonas vesicularibs strain 266XY4,which was identified as Brevundimonas vesiculari.Drug-resistant genetic detecting results showed that the strain has Sul1,aac(6′)-Ⅰb and TEM drug-resistant genes.The Kirby-Bane showed that the bacteria were resistant to piperacillin,kanamycin,ampicillin and so on.The results of drug susceptibility test were basically consistent with those drug resistance gene test.In this study,The strain was identified as Brevundimonas vesicularis,and was resistant to sulfonamide,aminoglycoside andβ-lactam antimicrobials,providing reference for the study of the resistance mechanism of Brevundimonas vesicularis.
