摘要
目的:构建适宜牙髓再生的透明质酸(HA)微球支架,评价其作为细胞载体的可行性。方法:乳化交联法制备透明质酸微球,SEM观察其形貌特征及粒径大小,将该微球材料与人牙髓干细胞(hDPSCs)共培养,MTT实验检测hDPSCs在微球存在的微环境内的增殖情况。结果:乳化交联法可制备透明质酸微球,该微球规则、圆整、粒径5~10μm;透明质酸微球浓度0.05~0.8 mg/ml作用24~72 h不抑制hDPSCs增殖,细胞毒性为0级。结论:透明质酸微球可作为应用hDPSCs的载体。
Objective:To construct hyaluronic acid(HA)microspheres scaffold suitable and to evaluate its citotoxicity to human dental pulp stem cells(hDPSCs).Methods:Hyaluronic acid microspheres were prepared by emulsification-cross-linking method,and their morphology and particle size were observed by SEM.The microspheres were co-cultured with hDPSCs,the proliferation of hDPSCs in the microspheres were evaluated MTT test.Results:By emulsification-cross-linking method,HA microspheres were prepared.The microspheres were regular and round with the diameter of 5-10μm.HA microspheres at 0.05-0.8 mg/ml showed no oytotoxicity to hDPSCs and can promote the proliferation of DPSCs.Conclusion:Hyaluronic acid microspheres can be used as DPSCs scaffold for pulp regeneration.
