摘要
目的:系统评价环状RNA(circRNA)在中国人群结直肠癌中的诊断价值。方法:在PubMed、Embase、Cochrane Library、Web of Science、CNKI、CBM、万方、维普等数据库检索,由两名研究人员按照纳入与排除标准,筛选2020年3月8日前发表的文献并提取数据。采用Review Manager 5.3、Meta-Disc 1.4、STATA 16.0进行Meta分析,计算其合并的灵敏度、特异度、阳性似然比、阴性似然比、诊断比值比和汇总受试工作者特征曲线(SROC)的曲线下面积(AUC)。结果:共纳入14篇文献,包括1 137例结直肠癌患者和915例非癌对照。合并的灵敏度、特异度、阳性似然比、阴性似然比、诊断比值比和AUC分别为:0.81[95%CI(0.74,0.86)]、0.72[95%CI(0.65,0.78)]、2.91[95%CI(2.35,3.59)]、0.27[95%CI(0.21,0.35)]、10.79[95%CI(7.59,15.34)]和0.83[95%CI(0.80,0.86)]。亚组分析结果显示环状RNA表达水平、样本量大小、样本类型是异质性的主要来源。结论:环状RNA可成为临床诊断结直肠癌的新型潜在生物标志物。
Objective:To conduct a systematic review on diagnostic value of circular RNA(circRNA)in colorectal cancer in Chinese population.Methods:Two investigators were designated to search in PubMed,Embase,Cochrane Library,Web of Science,CNKI,CBM,Wanfang,and VIP databases for literatures published up to March 8,2020,and complete data extraction according to inclusion and exclusion criteria.A meta-analysis was performed using Review Manager 5.3,Meta-Disc 1.4,STATA 16.0 softwares.Pooled sensitivity,specificity,positive likelihood ratio(PLR),negative likelihood ratio(NLR),diagnostic odds ratio(DOR)and area under the curve(AUC)of receiver operating characteristic curve(ROC)were calculated.Results:A total of 14 articles were included,covering 1137 patients with colorectal cancer and 915 non-cancer controls.The pooled sensitivity,specificity,PLR,NLR,DOR and AUC were 0.81[95%CI(0.74,0.86)],0.72[95%CI(0.65,0.78)],2.91[95%CI(2.35,3.59)],0.27[95%CI(0.21,0.35)],10.79[95%CI(7.59,15.34)]and 0.83[95%CI(0.80,0.86)].Subgroup analysis showed that the expression level of cirRNA,sample size,and sample type were the main sources of heterogeneity.Conclusion:CirRNA can be a novel potential biomarker for clinical diagnosis of colorectal cancer.
