利用高通量测序技术对水稻秸秆中、低温降解菌系的比较分析 被引量:6

Comparative Analysis of Rice Straw-Decomposing Mesophilic and Cryogenic Microbial Consortia Using High-Throughput Sequencing Technology

摘要 采用Illumina MiSeq高通量测序技术,对中、低温(中温25℃、低温10℃)富集驯化所得水稻秸秆降解菌系进行测序;采用生物信息学方法对中、低温秸秆降解菌系群落生物多样性进行分析。结果表明,测序质控后获得601489条16S rDNA序列,平均长度273 bp,经Silval 132数据库比对,中温降解菌系包括18个门,172个属,302个OTU,其中优势菌属为弓形杆菌属(Arcobacter)、拟杆菌属(Bacteroides)、屠场杆菌属(Macellibacteroides)、假单胞菌属(Pseudomonas)和梭形杆菌属(Lysinibacillus);低温降解菌系包括16个门,169个属,280个OTU,其中优势菌属为弓形杆菌属、屠场杆菌属、拟杆菌属、丛毛单胞属(Comamonas)和假单胞菌属。温度对降解菌系中优势菌门的相对丰度无显著影响,中、低温秸秆降解菌系主要菌属相对丰度差异显著。 Illumina MiSeq high-throughput sequencing technology was used to carry out sequencing for rice straw-decomposing microbial consortia obtained by enrichment and cultivation mesothermally and cryogenically(medium temp at 25℃,low temp at 10℃).Bioinformatics method was adopted to analyze the biodiversity of mesophilic and cryogenic straw-degrading consortia.The results showed that,the average length of sequencing was 273 bp,and a total of 601489 high-quality 16S rDNA sequences after quality control.These sequences were then compared with Silval 132 database.The sequences of mesophilic decomposing microbial consortia belonged to 18 phyla,172 genera and 302 OTUs,among them five dominant genera were Arcobacter,Bacteroides,Macellibacteroides,Pseudomonas and Lysinibacillus;and the cryogenic one belonged to 16 phyla,169 genera and 280 OTUs,among them five dominant genera were Arcobacter,Macellibacteroides,Bacteroides,Comamonas and Pseudomonas.Temperature had no significant effect on the relative abundance of the dominant phyla of decomposing microbial consortia.The main genera relative abundance was significantly different between mesophilic decomposing microbial consortia and the cryogenic one.
